龙牡桂枝汤对ADHD模型大鼠左右额叶-纹状体去甲肾上腺素转运体蛋白表达的影响 被引量:1

摘要 目的观察龙牡桂枝汤对注意缺陷多动障碍(ADHD)模型动物—幼年自发性高血压大鼠(SHR)左右额叶-纹状体去甲肾上腺素转运体(NET)蛋白表达的影响,探讨该药治疗ADHD的作用机制。方法采用幼年雄性SHR大鼠48只,随机分为龙牡桂枝汤高、中剂量组、盐酸哌甲酯对照组和生理盐水对照组,连续灌胃给药14天,采用Western-blot法检测各组大鼠左右脑前额叶皮质、纹状体NET蛋白表达水平。结果用药后,盐酸哌甲酯对照组、龙牡桂枝汤中、高剂量组大鼠右脑前额叶皮质和纹状体的NET蛋白表达增加,而各实验组大鼠左脑前额叶皮质和纹状体NET蛋白表达无明显变化。结论龙牡桂枝汤治疗ADHD的作用机制之一与其能上调右脑前额叶皮质、纹状体的NET蛋白表达有关。 Objective : To observe the effect of Longmu Guizhi Decoction on the left and fight prefrontal cortex and striatum norepinephrine transporter(NET) protein expression of attention deficit hyperactivity disorder(ADHD) animal models, i.e. young spontaneously hypertensive rats (SHR), and study the effect of the drug on ADHD mechanism. Methods: 48 young male SHR were randomly divided into Longmu phenidate group and a normal saline group. Each group was given blot was used to detect the NET protein levels of prefrontal cortex Guizhi high and medium dose groups, a methyl- intragastric administration for 14 days. Western - and striatum of the rats. Results: After the treat- ment, the right prefrontal cortex and striatum NET protein expression of the methylphenidate group, Longmu Guizhi high and medium dose groups increased, but the left prefrontal cortex and striatum NET protein expression of each test group had no significantly changes. Conclusion : One of the Longmu Guizhi Decoction mechanisms for ADHD treatment is related to its adjustment of the right prefrontal cortex and striatum NET protein expression.
