卫气虚证大鼠模型的建立及玉屏风散的反证效果 被引量:10

Modeling Rat with Weiqi Deficiency Syndrome and Disprooving Effect of Yupingfeng San(玉屏风散)

摘要 目的探索卫气虚证动物模型的建立方法并观察玉屏风散对该模型的反证效果。方法将50只SD大鼠随机分为正常对照组、环磷酰胺组、寒热刺激组、疲劳寒热组、中药干预组,每组10只。正常对照组常规饲养;环磷酰胺组腹腔注射环磷酰胺10 mg/kg,每日1次;寒热刺激组寒热交替刺激造模,每日1次;疲劳寒热组以疲劳联合寒热交替法造模;中药干预组造模方法同疲劳寒热组,每日造模后灌服玉屏风散颗粒25 g/kg,每日1次。各组均连续7天。观察各组大鼠一般情况及行为学表现,判断各造模方法与中医卫气虚证的相似度。比较各组大鼠体重、胸腺、脾脏指数、体外巨噬细胞吞噬功能、脾脏白细胞介素2(IL-2)、白细胞介素6(IL-6)水平。结果通过对各组大鼠行为学观察发现,疲劳寒热组大鼠模型更接近中医卫气虚证的宏观证候评定标准。与正常对照组比较,环磷酰胺组、疲劳寒热组、中药干预组大鼠体重显著降低(P<0.05﹚;与寒热刺激组比较,疲劳寒热组体重显著降低(P<0.05﹚;与疲劳寒热组比较,中药干预组体重显著升高(P<0.05﹚。与正常对照组比较,其余各组胸腺、脾脏指数,巨噬细胞吞噬率及吞噬指数,脾脏IL-2、IL-6水平均显著降低(P<0.05﹚;与疲劳寒热组比较,中药干预组胸腺、脾脏指数,巨噬细胞吞噬率及吞噬指数,脾脏IL-2、IL-6水平均显著升高(P<0.05﹚。结论疲劳联合寒热交替法造模是较好的中医卫气虚证大鼠模型造模方法,可被玉屏风散反证。 Objective To explore the modeling methods of animal models of Defensive-qi deficiency syndrome,and to observe the disproof effect of Yupingfeng San on the rat models. Methods Fifty sprague-dawley rats were randomized into normal control group( group A),cyclophosphamide group( group B),cold and hot stimulation group(group C),fatigue with cold and hot stimulation group(group D),and Chinese Traditional Medicine group(group E),with ten rats in each group. In group A,rats were fed routinely; in group B,10 mg / kg of cyclophosphamide was intraperitoneal injected to the rats every day; cold and hot stimulation was given to the rats once a day in group C;and made rats swim to fatigue,and then gave cold and hot stimulation once daily to the rats in group D and group E.In addition,Yupingfeng San 25 g / kg was given to rats in group E once day. Each group was treated for seven days.After then general condition and behavior-expressing of rats were observed in each group,the similarity between the results by each modeling method and Weiqi deficiency syndrome was estimated,and the body weight,thymus index,spleen index,phagocytic function of extracorporeal macrophage,and spleen levels of interleukin-2( IL-2) and interleukin-6( IL-6) were compared between groups. Result It was found in behavior-expressing of rats that the model of fatigue with cold and hot stimulation in Group D was more close to the macroscopic evaluation standard of Weiqi deficiency syndrome. Compared with group A,body weight of rats reduced significantly in group B,D,and E( P 0. 05). Compared with group C,body weight of rats reduced significantly in group D( P 0. 05). Compared with group D,body weight of rats increased remarkably in group E( P 0. 05). In addition,compared with group A,thymus index and spleen index,phagocytic rate and phagocytic index of macrophage,and levels of IL-2 and IL-6reduced significantly in group B,C,D,and E( P 0. 05). Compared with group D,thymus index and spleen index,phagocytic rate and phagocytic index of macrophage,and levels of IL-2 and IL-6 increases significantly in group E( P 0. 05). Conclusion Fatigue with cold and hot stimulation is a preferable method to establish rat model with Weiqi deficiency syndrome. This conclusion can be disproved by Yupingfeng San.
