姜黄素对肺纤维化大鼠骨髓及外周血内皮祖细胞的影响 被引量:6

摘要 目的探讨姜黄素对肺纤维化大鼠骨髓及外周血中内皮祖细胞(EPCs)数量、内皮型一氧化氮合酶(e NOS)表达的影响及其机制。方法采用经气管灌注博莱霉素(BLM)的方法制作肺纤维化模型。60只大鼠完全随机分为对照组、模型组、姜黄素组。每组于第14天、28天各处死10只大鼠。肺组织行HE染色观察并进行肺纤维化Ashcroft评级,采用流式细胞术检测骨髓及外周血中EPCs的数量,Western blot法检测骨髓中e NOS蛋白的表达。结果姜黄素组肺泡炎和肺纤维化程度明显低于模型组。与对照组比较,模型组各时间点骨髓及外周血EPCs数量及e NOS蛋白的表达明显降低(P<0.01)。与模型组比较,姜黄素组各时间点骨髓及外周血EPCs数量及e NOS蛋白的表达亦明显增多(P<0.05)。结论姜黄素可上调肺纤维化大鼠骨髓及外周血EPCs数量,促进EPCs动员入血,其作用可能与姜黄素激活e NOS通路相关。 AIM To study the effect of curcumin on the number of endothelial progenitor cells,( EPCs) the protein expression of e NOS in bone marrow and peripheral blood in rats with pulmonary fibrosis and explore their possible mechanism. METHODS Pulmonary fibrosis model of SD rats was reproduced by the single intratracheal perfusion of bleomycin. Sixty rats were randomly divided into the control group,model group, and curcumin group. Ten rats were sacrificed from each group on the 14 thand 28thday. The lung tissue was observed by HE staining and evaluated for the formation of lung fibrosis by using Ashcroft scoring system. Flow cytometer was used to determine the percentage of EPCs in bone marrow and peripheral blood. The protein expression of e NOS was detected by Western-blot. RESULTS The alveolitis and the pulmonary fibrosis in the curcumin group were significantly alleviated. In comparison with the control group,the percentage of EPCs in bone marrow and peripheral blood and the protein expression of e NOS were significantly reduced in the model group( P〈 0. 01). In comparison with the model group,the percentage of EPCs in bone marrow and peripheral blood and the protein expression of e NOS were significantly up-regulated in curcumin group( P〈 0. 05). CONCLUSIONS Curcumin can effectively upregulate the percentage of EPCs in bone marrow and peripheral blood,and help bone marrow EPCs migrate into peripheral blood through activating e NOS pathway in rats with pulmonary fibrosis.
