摘要
目的建立HPLC法同时测定五味消毒饮中绿原酸、咖啡酸、蒙花苷和秦皮乙素4种活性成分。方法五味消毒饮水提液分析采用Agilent Eclipse XDB-C18色谱柱(250 mm×4.6 mm,5.0μm),以0.05%三氟乙酸溶液(V/V)-乙腈为流动相,梯度洗脱,体积流量1.0 m L/min,检测波长为320 nm和344 nm,柱温30℃。结果绿原酸、咖啡酸、蒙花苷和秦皮乙素的线性范围分别为4.96-49.6μg/m L,0.255-2.55μg/m L,2.51-25.1μg/m L和2.47-24.7μg/m L(r≥0.999 8)。平均回收率在95%-105%范围内。结论该方法简便、灵敏、准确,可作为五味消毒饮的质量控制方法。
AIM To establish an HPLC method for simultaneously determining chlorogenic acid,caffeic acid,linarin,and aesculetin from Wuwei Xiaodu Drink. METHODS The analysis of aqueous extract of Wuwei Xiaodu Drink was performed on Agilent Eclipse XDB-C18column( 250 mm × 4. 6 mm,5. 0 μm) with mobile phase consisting of 0. 05%( V / V) trifluoroacetic acid-acetonitrile in a gradient elution mode. The wavelength was set at320 nm and 344 nm for different components. Flow rate was 1. 0 m L / min and column temperature was maintaned at30 ℃. RESULTS The linear ranges of chlorogenic acid,caffeic acid,linarin,and aesculetin were 4. 96-49. 6 μg / m L,0. 255-2. 55 μg / m L,2. 51-25. 1 μg / m L,and 2. 47-24. 7 μg / m L( r≥0. 999 8),respectively. The average recoveries were between 95% - 105%. CONLUSION The method is simple,sensible,accurate and can be used for the quality control of Wuwei Xiaodu Drink.
