Alterations of plasmacytoid dendritic cells in peripheral blood of patients with primary Sjogren′s syndrome and their significance

摘要 目的:探讨原发性干燥综合征( primary Sj?gren′s syndrome,pSS)患者外周血浆细胞样树突状细胞pDC( Lin-CD123+HLA-DR+)的变化及意义。方法收集疾病组原发性干燥综合征患者39例,正常对照组15例,采用流式细胞术检测pDC占外周血淋巴细胞的比例,ELISA检测15例原发性干燥综合征患者血浆IFN-α、IL-6和TNF-α水平。结果 pSS疾病组外周血pDC占淋巴细胞百分比低于正常对照组(P<0.05);抗干燥综合征抗原A(SSA)抗体阳性pSS患者pDC水平显著高于抗SSA抗体阴性pSS患者(P<0.05);pSS疾病组血浆TNF-α(P<0.05)和IL-6(P<0.05)水平高于正常对照组,pSS疾病组血浆IFN-α水平与正常对照组相比差异无统计学意义( P>0.05);pSS疾病组外周血pDC占淋巴细胞百分比与血浆TNF-α、IL-6和IFN-α水平之间无相关性。结论 pSS患者外周血pDC占淋巴细胞百分比降低,外周血TNF-α、IL-6水平增高,可能参与原发性干燥综合征的发病机制。 Objective To investigate the alterations of plasmacytoid dendritic cells ( pDC) in pa-tients with primary Sj?gren′s syndrome ( pSS) and their significance.Methods Peripheral blood samples were collected from 39 patients with pSS and 15 healthy subjects.The percentages of pDC ( Lin-CD123+HLA-DR+) in peripheral blood samples were analyzed by flow cytometry.The levels of IFN-α, TNF-αand IL-6 in serum samples were detected by ELISA.Results The percentages of pDC in patients with pSS were lower than those in healthy subjects (P〈0.05).Higher levels of pDC were detected in patients positive for anti-Sj?gren syndrome antigen A ( anti-SSA) antibody as compared with those in patients negative for anti-SSA antibody.The levels of TNF-αand IL-6 in serum samples from patients with pSS were higher than those in healthy subjects.Conclusion Patients with pSS showed decreased percentages of pDC population, but increased levels of TNF-αand IL-6.The changes of pDC population, TNF-αand IL-6 in peripheral blood might play active roles in the pathogenesis of pSS.
