摘要
目的:探讨MALAT-1对胃癌细胞系SGC7901侵袭和迁移能力的影响。方法:用实时定量聚合酶链反应(RT-PCR)的方法在mRNA水平测定MALAT-1在20对胃癌组织及癌旁正常组织的表达,以及在GES和SGC7901细胞中的表达。RNA干扰(RNAi)技术下调胃癌细胞系SGC7901中MALAT-1的表达后,以高内涵细胞运动试验检测其运动能力,Transwell实验检测其侵袭和迁移能力。结果:RT-PCR结果显示MALAT-1在胃癌组织中的表达高于癌旁正常组织;相比GES,MALAT-1在SGC7901中的表达升高。下调MALAT-1在SGC7901中的表达后,高内涵细胞运动试验和Transwell实验结果显示SGC7901的运动、侵袭和迁移能力均明显减弱。结论:MALAT-1能够促进胃癌细胞SGC7901的侵袭和迁移能力。
Objective: To investigate the influence of MALAT- 1 on the invasion and migration in human gastric cancer cell line SGC7901. Methods: The mRNA levels of MALAT- 1 in 20 pairs of the gastric cancer tissues and adjacent normal tissues,GES and SGC7901 were determined by real- time PCR( RT- PCR) assays. The influence of MALAT- 1 on the mobility,invasion and migration of gastric cancer cell line SGC7901 were detected using high content analysis and Transwell assay. Results: The results of RT- PCR showed that MALAT- 1 levels were up- regulated in gastric cancer tissues compared with adjacent normal tissues,and the expression level of MALAT- 1 in SGC7901 was significantly higher than that in GES. High content analysis and Transwell assay showed that the mobile,invasive and migratory ability were decreased after down- regulated the expression of MALAT- 1 in gastric cancer cell line SGC7901. Conclusion: MALAT- 1 could enhance the invasive and migratory ability of gastric cancer cell SGC7901.
