摘要
[目的]评价注射用重组人Ⅱ型TNF受体-抗体融合蛋白(tumour necrosis factor FC fusion protein,rh TNFR:Fc)(商品名:益赛普)在兔正畸微种植体周围炎模型的作用。[方法]选用新西兰兔12只,随机分为模型对照组和治疗组。分别在兔下颌无牙区植入正畸微种植体,采用钢丝结扎法建立实验性微种植体周围炎模型,结扎56d,治疗组给予rh TNFR:Fc,剂量1.1mg mg/(kg·d)1周,模型对照组给予等量生理盐水,两组在造模后第14、56、63d检测改良菌斑指数(m PLI)、改良龈沟出血指数(m BI)和种植体周探诊深度(PD);造模后63d取种植体周围牙龈行组织病理学检查,常规HE、免疫组化染色,观察炎性细胞浸润及TNF-α分布情况。[结果]造模后63d,治疗组m PLI、m BI、PD较模型对照组均明显下降(P<0.05)。组织病理学检查显示:造模后63d治疗组牙龈组织内炎症细胞浸润、TNF-α均较模型对照组减少(P<0.05)。[结论]rh TNFR:Fc可影响微种植体周围炎炎症反应过程。
[Objective] The aim of the study was to evaluate the effect of recombinant human tumour necrosis factor FC fusion protein(rhTNFR:Fc) on the non-surgical treatment peri-implantitis in a rabbit modeL[Methods] 12 male adult rabbits were randomly chosen and divided into control group and treatment group. Peri-implantitis of micro implant in rabbit model was induced by securing silk ligatures around the implants in mandibular toothless alveolar for 8 weeks. Treatment group received rhTNFR:Fc(1.1mg·kg^-1 daily for 1 week) and control group received saline. Modified plaque and calculus index(mPLI),modified gingival blooding index(mBI),probing depths(PD) were evaluated before and after the application of rhTNFR: Fc.The level of inflannnatory cell infiltration and tumor necrosis factor-u in gingival tissues were measured. [Result] Compared with control group, mPLI, mBI and PD of the treatment group were significantly decreased after application of rhTNFR:Fc(P〈0.05), inflammatory cell infiltration and TNF-α in treatment group was significantly lower(P〈0.05). [Conclusion] rhTNFR:Fc could influence the course of the inflammatory response in peri-implantitis and might contribute to the treatment of peri-implantitis.
