妊娠期高血压患者胎盘中IL-2和MMP-9表达水平的实验研究

Experimental study on the expression levels of IL-2 and MMP-9 in placenta in patients with hypertensive disorder complicating pregnancy

目的:通过观察妊娠期高血压患者胎盘中IL-2和MMP-9的表达水平探讨妊娠期高血压疾病的发病机制。方法:选择妊娠期高血压孕妇114例,其中38例为轻度、35例为中度、41例为重度;另选择20例正常孕妇作为对照组,4组均于胎盘娩出后立即在胎盘脐带的对侧剪取胎盘组织,采用实时荧光定量PCR方法和酶联免疫法检测胎盘组织中IL-2和MMP-9的mRNA水平和蛋白含量。结果:轻度组孕妇与对照组孕妇胎盘组织中IL-2、MMP-9的mRNA水平及蛋白含量比较差异无统计学意义(P>0.05);中、重度组孕妇MMP-9的mRNA水平及蛋白含量均低于对照组孕妇,IL-2的mRNA水平及蛋白含量均高于对照组孕妇,差异均有统计学意义(P<0.01)。结论:妊娠期高血压孕妇胎盘组织中IL-2、MMP-9的mRNA水平及蛋白含量随着分级增高而变化,为临床治疗妊娠期高血压疾病提供新的靶点。

Objective： To explore the pathogenesis of hypertensive disorder complicating pregnancy（ HDCP） by observing the expression levels of interleukin- 2（ IL- 2） and matrix metalloproteinase- 9（ MMP- 9） in placenta in HDCP patients. Methods： A total of114 patients with HDCP were selected,including 38 patients with mild HDCP,35 patients with moderate HDCP,and 41 patients with severe HDCP; 20 normal pregnant women were selected as control group. Placental specimens were obtained after delivery of placenta in the four groups; real- time fluorescent quantitative PCR and ELISA were used to detect the levels of IL- 2 and MMP- 9 mRNA and proteins in placental tissues. Results： There was no statistically significant difference in the levels of IL- 2 and MMP- 9 mRNA and proteins in placental tissues between mild HDCP group and control group（ P 〈0. 05）; the levels of MMP- 9 mRNA and protein in moderate HDCP group and severe HDCP group were statistically significantly lower than those in control group（ P 〈0. 01）; the levels of IL- 2 mRNA and protein in moderate HDCP group and severe HDCP group were statistically significantly higher than those in control group（ P 〈0. 01）. Conclusion： The levels of IL- 2 and MMP- 9 mRNA and proteins in placental tissue of HDCP patients change with the increase of grading,which provides a new target for clinical treatment of HDCP.
