老年、高龄及长寿老人肝脾组织基因组DNA含量的变化及比较 被引量:1

摘要 目的观察尸检老年、高龄及长寿老人肝、脾组织基因组DNA含量的变化及差异,探讨衰老进程中DNA含量的变化规律并为今后的衰老相关研究提供实验数据。方法选取24 h内老年尸检(≥65岁)低温(-80℃)保存的肝组织35例和脾组织33例,按照年龄分为3组:65~79岁组、80~89岁组、≥90岁组(其中包括百岁老人2例);低温(-80℃)保存的肝、脾组织用试剂盒提取基因组DNA,紫外光谱法测定肝脾组织DNA含量,并进行比较分析。结果肝组织DNA含量:与65~79岁组(1.464±0.488)mg/m L比较,80~89岁组(0.310±0.286)mg/m L显著降低(P〈0.05);且≥90岁组(1.147±0.333)mg/m L的DNA含量显著高于80~89岁组(P〈0.05)。脾组织基因组DNA含量:与65~79岁组(1.723±0.726)mg/m L比较,80~89岁组(0.938±0.589)mg/m L显著降低(P〈0.05);且≥90岁组(1.688±0.963)mg/m L的DNA含量显著高于80~89岁组(P〈0.05)。同一尸检者肝、脾组织DNA含量比较:肝组织平均DNA含量(0.856±0.658)mg/m L显著低于脾组织平均DNA含量(1.414±0.852)mg/m L(P〈0.05)。结论老年人肝、脾组织基因组DNA含量随增龄而降低;90岁以上肝、脾组织DNA的含量均有所增加;同一尸检者肝组织DNA含量显著低于脾组织DNA含量,提示不同脏器组织DNA含量存在差异。 Objective To investigate the change of genomic DNA of liver and spleen tissue for different age of the elderly,and provide the experimental data for aging-related research. Methods 35 livers and 33 spleens of autopsied samples preserved in refrigerator at- 80 ℃ were divided into 3 groups according to age:age 65 y to 79 y,age 80 y to 89 y,age≥90y. The content of DNA in liver and spleen was determined by ultraviolet absorbent method. Results Compaired with age 80 y to 89y(0. 310 ± 0. 286) mg / m L,the content of DNA in liver was significant higher at age 65 y to 79y(1. 464 ± 0. 488) mg / m L and age ≥ 90y(1. 147 ± 0. 333) mg / m L( P〈0. 05);Compared with age80 y to 89y(0. 938 ± 0. 589) mg / m L,the content of DNA in spleen was significant higher at age 65 y to 79y(1. 723 ± 0. 726) mg / m L and age≥90y(1. 688 ± 0. 963)mg/m L(P〈0. 05). The content of DNA was significant lower in liver(0. 856 ± 0. 658)mg/m L than that in spleen(1. 414 ± 0. 852)mg/m L. Conclusion The content of DNA in human liver and spleen tissue may be decrease along with aging. The content of DNA in the group at age≥90y may be increase. There were some differences between different viscera tissue in content of DNA.
