摘要
[目的]利用生物信息学方法分析斑马鱼过氧化物还原酶Peroxiredoxin(Prx)家族的结构和功能。[方法]基于NCBI数据库中的BLAST程序,搜索斑马鱼Prx序列,从蛋白质氨基酸序列比对、系统发育、信号肽、跨膜区、二级结构、高级结构及蛋白质相互作用等方面进行解析。[结果]斑马鱼Prx家族有6个成员(Prx1-Prx6),均含有抗氧化活性半胱氨酸Cys,不含有跨膜结构域,仅Prx4含有信号肽。Prx1-Prx4序列同源性大于50%,有共同的祖先。Prx1-Prx5可与硫氧还蛋白相互作用,Prx6可与谷胱甘肽相互作用。脯氨酸、苏氨酸和精氨酸在空间结构上与斑马鱼Prx活性半胱氨酸接近,有利于Prx抗氧化活性发挥。[结论]斑马鱼Prx蛋白结构具有脊椎动物Prx特点,具有潜在的抗氧化生物学活性。
[Objective]To analyze the structure and function of peroxiredoxin family of Danio rerio using bioinformatics methods. [Methods]Based on the BLAST program of NCBI database,the peroxiredoxin sequence of Danio rerio was searched. The amino acid sequence alignment,signal peptide,transmembrane region,phylogenesis,secondary structure,tertiary structure and protein-protein interaction were researched by using bioinformatics analysis. [Results]Six Prxs(Prx1-Prx6) were found in Danio rerio. All the Prxs had active cysteine,and only Prx4 had signal peptide. Prx1 to Prx4 showed more than 50% sequence homologous with each other,which indicated that they had common ancestor. Prx1 to Prx5 used thioredoxin as the electron donor and Prx 6 interacted with glutathione. The conserved amino acids proline,threonine and arginine were closed to the N-terminal cysteine which contributed to the antioxidative activity of Prx. [Conclusion]Consistent with the characteristic feature of vertebrate Prx,Prxs of Danio rerio also had the potential activity of antioxidant.
