摘要
目的观察E-cadherin蛋白及其mRNA在喉鳞状细胞癌的表达活性。方法原发性喉鳞状细胞癌30例,手术中同时留取每例的瘤组织标本及癌旁组织标本,分别以qRT-PCR检测其E-cadherin mRNA表达量、免疫组化法检测E-cadherin蛋白表达水平,比较分析其间的表达水平差异。结果免疫组化显示,癌旁组织E-cadherin平均分值为2.51±0.96,喉癌组织则为1.57±0.73,癌旁组织的表达量明显高于喉癌组织(t=8.14,P<0.05);qRT-PCR检测结果显示,癌旁组织E-cadherin mRNA表达水平为4.58±1.96,喉癌组织为2.51±1.03,前者表达量明显高于后者(t=6.26,P<0.05)。结论 E-cadherin活性变化可能涉及喉鳞状细胞癌的进展,可以作为诊断喉鳞状细胞癌的潜在标记物。
Objective To investigate the expressive Methods The expressive activities of E-cadherin in activity of E-cadherin in laryngeal squamous cell carcinoma. cancerous tissue samples and their adjacent non-neoplastic tissues from patients with primary laryngeal squamous cell carcinoma were determined by immunohistochemistry and qRT-PCR assay to detect both E-cadherin protein and its mRNA respectively. Then, the data were analyzed carefully to test their expressive differences and possible clinical implications. Results As shown from immunohistochemistry, the scores of E-cadherin in the adjacent non-neoplastic tissue samples was 2.51 ±0.96, while it was 1.57±0.73 in cancerous tissue samples, with obviously higher value in the former than in the latter (t=8.14, P〈0.05). Meanwhile, the expressive level of E-cadherin mRNA was 4.58±1.96in the adjacent nonneoplastic tissue samples and it was 2.5 ±1.03 in cancerous tissue samples, also significantly higher in the adjacent non-neoplastic tissue samples (t=6.26, P〈0.05). Conclusions Changes in the expressive activity of E- cadherin may be involved in the progression of potential diagnostic marker for such a lesion. laryngeal squamous cell carcinoma and it can be taken as potential diagnostic marker for such a lesion.
