摘要
建立了同时测定藏中当药黄素、当药醇苷、苦龙苷含量的方法。采用反相高效液相色谱法(RPHPLC),以Kromasil-C18(250 mm×4.6 mm,5μm)为色谱柱,以甲醇(A)-0.04%磷酸溶液(B)为流动相,梯度洗脱,40%A(0 min)-63%A(20 min);流速为0.8 m L/min;柱温为30℃,检测波长为254 nm。结果表明,在20 min内完成对3种成分的分析,各成分之间均达到基线分离。当药黄素、当药醇苷、苦龙苷的线性范围分别为:0.675 0~4.050 0μg(R2=0.999 5)、0.005 5~0.330 0μg(R2=0.999 5)、0.027 5~0.165 0μg(R2=0.999 5),线性关系良好。平均加样回收率(n=9)分别为99.90%、99.15%、99.88%。建立的HPLC分析方法简单、准确,能快速测定藏药肋柱花中有效成分的含量。
RP-HPLC was used to determine the content of swertisin, swertianolin, amarogerth in Tibetan herb. The sample was separated on the column of Kromasil-C18(250 mm×4.6 mm,5 μm) eluted with methanol(A) and 0.04% phosphoric acid solution(B). The initial condition was 40%A, then rose to 63% A in 20 min with detection wavelength at 254 nm, flow rate0.8 m L / min, and column temperature 30 ℃. And three active constituents were base-isolated. The linear ranges of swertisin,swertianolin, amarogerth, were 0.675 0~4.050 0 μg(R2=0.999 5), 0.005 5~0.330 0 μg(R2=0.999 5), and 0.027 5~0.165 0 μg(R2=0.999 5)respectively; the average recoveries(n=9) were 99.90%, 99.15% and 99.88% respectively. The method was easy and precise which can be used for content analysis of active constituents in Lomatogonium rotatum(L.)Fries es Nym rapidly.
