摘要
目的:应用RNA干扰技术,靶向敲低人胃癌MGC-803细胞膜联蛋白A7(Annexin A7,ANXA7)的表达,观察ANXA7低表达对MGC-803细胞增殖的影响。方法:实验分为3组,以靶向ANXA7的si RNA转染MGC-803细胞为si RNA干扰组,另设阴性对照组和空白对照组,以Western blot法检测转染后MGC-803细胞ANXA7的表达,应用MTT法、克隆形成实验检测ANXA7低表达对MGC-803细胞的增殖能力的影响。结果:si RNA干扰组细胞ANXA7的表达较阴性对照组和空白对照组明显降低(P<0.05)。敲低ANXA7表达后,MGC-803细胞的增殖活性和克隆形成能力明显降低(P<0.05)。结论:ANXA7低表达可抑制胃癌MGC-803细胞的增殖。
Objective:To investigate low expression of Annexin A7(ANXA7) on proliferation of gastric cancer MGC-803 cells by target-silencing ANXA7 expression using RNA interference technique.Methods:MGC-803 cells were grouped into si RNA interference group, negative control group and blank control group. RNA interference technology was used to transfect the si RNA targeted ANXA7 into MGC-803 cells to knock down expression of ANXA7. Western blot was used to detect the expression of ANXA7 after transfection; MMT assay and clone formation assay to detect the effects of low expression of ANXA7 on proliferation of MGC-803 cells. Results:The expression of ANXA7 in MGC-803 cells of si RNA interference group were obviously lower than negative control group and blank control group(P〈0.05). The proliferation activity and colony forming ability of MGC-803 cells declined after knocking down expression of ANXA7(P〈0.05). Conclusions:Low expression of ANXA7 can inhibit proliferation of gastric cancer MGC-803 cells.
