宫颈癌组织miR-574-5p的表达及其下调后对宫颈癌SiHa细胞的影响 被引量:14

摘要 目的探讨宫颈癌患者组织中miR-574-5p的表达及其下调时对宫颈癌Si Ha细胞增殖、凋亡和迁移的影响。方法用实时荧光定量PCR测定80例宫颈癌组织中miR-574-5p水平。将miR-574-5p抑制物转染宫颈癌Si Ha细胞,四甲基偶氮唑盐比色法(MTT法)检测细胞增殖能力,流式细胞仪检测细胞凋亡率,Tanswell迁移实验检测细胞侵袭能力。结果宫颈癌组织中miR-574-5p相对表达水平高于正常宫颈组织(P<0.05);高表达的miR-574-5p与肿块大小、临床分期、病理分级和淋巴结转移有关(P<0.05);转染miR-574-5p抑制物组与空白组及阴性对照组相比,Si Ha细胞增殖能力下降(P<0.05);细胞凋亡率明显增加(P<0.05);转染后细胞穿膜能力明显减弱(P<0.05)。结论 miR-574-5p的高表达与宫颈癌的进展有关,下调miR-574-5p的表达抑制了宫颈癌Si Ha细胞的增殖和迁移,促进其凋亡,有望成为宫颈癌基因治疗的靶点。 Objective To investigate the expression level miR-574-5p on SiHa ceils. Methods The level of tissue samples as controls were detected by real-time RT-PCR. of tissue miR-574-5p and the effects of down-regulation of miR-574-5p in 80 samples cervical cancer and 60 normal Transfected the miR-574-5p inhibitor into SiHa cells, and detected the proliferation, the apoptotic rate and the migration of SiHa cell through MTI', the flow cytometer and Transwell experiment respectively. Results Expression levels of tissue miR-574-5p were significantly higher in cervical cancer than that in normal controls( P 〈 0. 001 ) ; Up-regulation expression of miR-574-5p had a sig- nificantly positive correlation with the International Federation of Gynecology and Obstetrics (FIGO) stage and pathological grade and lymph node metastasis (P 〈 0.05 ) , while it had no clear-cut correlations with the age of patients and menopausal status. After transfected the miR-574-5p inhibitor into SiHa cells, the proliferation and the migration were decline (P 〈 0. 05 ), and the apoptotic rate was raised (P 〈 0.05 ). Conclusions The highexpression of miR-574-5p in the tissue of cervical cancer may be related to the progress in the malignant cervical cancer, which is then expected to be a potential therapy of cervical cancer through the effects of transfecting miR- 574-5p inhibitor on SiHa cells.
