摘要
目的:确定海藻糖对过氧化氢诱导的M14黑素瘤细胞损伤的保护作用。方法:不同浓度过氧化氢处理后的M14黑素瘤细胞加入0.5μg/mL、1μg/mL、10μg/mL海藻糖,采用MTT法检测各组细胞存活率,嘌呤氧化酶法检测处理前后超氧化物歧化酶(SOD)的活性。结果:浓度为1μg/mL的海藻糖抗氧化作用明显,可提高低浓度过氧化氢(小于400μmol/L)损伤后的M14黑素瘤细胞存活率约33%,可提高SOD活力约20%。当过氧化氢浓度大于400μmol/L时,各浓度海藻糖对提高细胞存活率无明显作用。结论:1μg/mL海藻糖对低浓度(小于400μmol/L)过氧化氢诱导的M14黑素瘤细胞损伤具有保护作用。
Objective:To determine the protective effect of trehalose against the damage of hydrogen peroxide to the melanoma cell lines M14. Methods:Melanoma cell lines M14 induced by hydrogen peroxide were recultured by different concentrations of trehalose (0.5 μg/mL, 1 μg/mL, 10 μg/mL). The cell survival rate was detected by MTT. The activity of SOD was detected by xanthine oxidase technique. Results:The an-tioxidation of 1μg/mL trehalose was obvious. 1μg/mL trehalose improved the survival rate and the activity of SOD of M14 cells induced by low-concentration (〈400μmol/L) hydrogen peroxide by 33% and 20% respec-tively. The survival rate of M14 cells induced by high-concentration (〉400 μmol/L) hydrogen peroxide was not increased after re-cultured in different concentrations of trehalose. Conclusion:1μg/mL trehalose is pro-tective against the damage of low-concentration (〈400 μmol/L ) hydrogen peroxide to the melanoma cell lines M14.
