肌细胞增强因子2A基因P279L变异对eNOS的影响

摘要 目的:探讨肌细胞增强因子2A基因P279L变异对内皮型一氧化氮合酶(eNOS)的影响。方法:选自西北地区385名汉族人,采用酚氯法提取全基因组DNA,酶联免疫吸附试验(ELISA)测定血清中eNOS。结果:在MEF2A-P279L的PP、PL、LL的3种基因型中,eNOS水平差异有统计学意,LL型eNOS水平明显低于LP型和PP型,PL型100.35±12.61pg/ml与PP型103.28±13.35pg/ml差异无统计学意义(P>0.05),但eNOS水平成上升趋势。结论:MEF2AP279L变异影响eNOS水平,L等位基因可能具有致动脉粥样硬化的作用。 Objectives:To investigate the relationship between the distribution of Myocyte enhancer factor2A(MEF2A)gene P279 Lpolymorphism and plasma level of eNOS in Han nationality in Chinese North-Western area.Methods:Three hundred and eighty five Han ethnic subjects.DNA was extracted by phenol and chloroform extraction.The serum of endothelial nitric oxide synthase(eNOS)was measured by enzyme-linked immunosorbent assay(ELISA).Results:The level of eNOS showed significant difference in the sequence of PP,PL and LL genotypes(P0.05),the levels of LL were significantly lower than those in the LP and PP,no significant difference between PL genotype(100.35±12.61pg/ml)and PP genotype(103.28±13.35pg/ml)(P0.05),but The level of eNOS between PL and PP showed an upward trend.Conclusion:MEF2A-P279 Lgenetic variation results in a protective change in level of eNOS,L allele may have induced atherosclerosis.
