高效液相色谱法测定鼻咽清毒剂中4种活性成分的含量

HPLC determination of 4 active ingredient content in Biyan Qingdu granules

目的建立鼻咽清毒剂中4种活性成分木犀草苷、蒙花苷、噻嗪二酮苷和3,5-O-二咖啡酰基奎宁酸的含量测定方法。方法依利特C18色谱柱（4.6 mm×250 mm,5μm）;流动相为甲醇-0.1%磷酸溶液,梯度洗脱,流速0.9 m L·min-1;检测波长334 nm（木犀草苷、蒙花苷）;254 nm（噻嗪二酮苷、3,5-O-二咖啡酰基奎宁酸）;柱温30℃。结果在该色谱条件下,木犀草苷、蒙花苷、噻嗪二酮苷和3,5-O-二咖啡酰基奎宁酸分离良好,质量浓度分别在9.84~196.80μg·m L-1（r=0.999 8）、11.92~238.40μg·m L-1（r=0.999 6）、6.85~137.00μg·m L-1（r=0.999 9）、5.60~112.00μg·m L-1（r=0.999 4）时与其峰面积呈良好的线性关系,加样回收率依次为96.9%、99.0%、97.8%、96.9%,RSD依次为1.4%、1.4%、1.6%、0.71%（n=6）。结论所建立的方法简便、准确、重复性好,可用于鼻咽清毒剂的质量控制。

Objective To determine the content of 4 ingredients in Biyan Qingdu granule was developed by HPLC method. Methods HPLC separation was performed on Hypersil C18 column（4.6 mm×250 mm, 5 μm） with a gradient elution method, 0.1% phosphoric acid solution was used as the mobile phase at 0.9 m L·min- 1. The column temperature was 30 ℃. The wavelenth was 334 nm for luteolin-7-O-glucoside, buddleoside and 254 nm for xanthiside,（-）-3, 5-dicaffeoyl quinic acid. Results Luteolin-7-O-glucoside, buddleoside, xanthiside and（-）-3, 5-dicaffeoyl quinic acid were baseline separated and have a good linearity at 9.84- 196.80 μg·m L- 1（r =0.999 8）, 11.92- 238.40 μg·m L- 1（r = 0.999 6）, 6.85- 137.00 μg·m L- 1（r = 0.999 9）, and 5.60- 112.00 μg·m L- 1（r = 0.999 4）, respectively. The average recoveries and RSDs were 96.9%（1.4%）, 99.0%（1.4%）, 97.8%（1.6%） and 96.9%（0.71%）, respectively. Conclusion The method is simple, accurate, and shows good precision and reproducibility, which can be used for the quality control of Biyan Qingdu granules.
