Etiological Study on Tembusu Virus and Pasteurella multocida from 6-day-old Chick

摘要 对一例疑似坦布苏病毒和多杀性巴氏杆菌混合感染的6日龄鸡进行病原的分离鉴定。用套氏RT-PCR方法和常规细菌分离鉴定方法进行病毒、细菌的分离、鉴定,并对其病原的主要生物学特性进行了研究。结果表明:病毒经鉴定为坦布苏病毒,与鹅源坦布苏病毒JS804株核苷酸同源性达99%;分离到的细菌为多杀性巴氏杆菌。细菌对鸡的回归试验显示,0.1mL的菌液可致20周龄SPF鸡死亡;对6周龄Balb/c小鼠的致病性试验结果表明,含约10 CFU的毒液即可在24h内致其4/4死亡。 By using RT-PCR and conventional test methods , the author isolated and identified the pathogens in a 6-day-old chick, which was suspected of being infected by Tembusu virus and Pasteurella multocida simultaneously, and studied the main bio-logical characteristics of these two pathogens .The isolated virus was identified as Tembusu virus , which had 99%nucleic acid se-quence homology with goose-derived Tembusu virus strain JS804 .The isolated bacterium was identified as Pasteurella multocida. The results of bacterial pathogenicity tests showed that injecting 0.1 mL P.m ultocida liquid could lead to the death of 20-week-old SPF chickens;injecting the bacterial liquid containing 10 CFUs could cause the whole death of four 6-week-old Balb/C mice within 24 hours.
