摘要 纳米孔单分子分析技术是通过不同的压力驱动分子通过纳米孔,监测分子易位时产生的电流变化。该技术快速,无需标记、扩增等方法,可以检测分析分子性质。随之发展起来的纳米孔测序技术可以将人类基因组测序成本控制在1000美元之内。主要介绍了用于单分子分析的纳米孔技术的研究进展。 Nanopore analysis is a new technique that involves using a voltage to drive molecules through a nanoscale pore in an insulating membrane and monitoring the current across the pore.This approach allows molecules to be analysed rapidly without the need for labels or amplification.Recent advances suggest that nanopore-based sensors could be competitive with other third-generation DNA sequencing technologies,and may be able to rapidly and reliably sequence the human genome for under$1000.The development of techniques for nanopore-based single-molecule analysis is reviewed.
