坎地沙坦对早期糖尿病大鼠视网膜中Akt蛋白及其磷酸化位点表达的影响

摘要 Akt作为磷脂酰肌醇3-激酶(P13K)信号通路的主要下游分子,在P13K或缺氧微环境的刺激下发生磷酸化,其中Thr308位点和Ser473位点的同时磷酸化为Akt活化所必需[1]。PI3K/Akt信号通路在糖尿病(DM)血管内皮炎症、凋亡、增生调控中起关键作用[2]。P13K/Akt激活剂坎地沙坦以其具有独立于降低血压以外的保护作用,已广泛应用于心脑血管及肾病的治疗[3-4]。但坎地沙坦是否可以通过激活PI3K/Akt信号通路进而参与对视网膜细胞的保护作用尚不清楚。为此,我们对一组早期糖尿病大鼠进行消化道灌注坎地沙坦,观察其对大鼠视网膜中Akt蛋白及其磷酸化位点AktSer473表达的影响。现将结果报道如下。
