摘要
目的比较3种屋尘螨提取液(house dust mite extract,HDM)对BALB/c小鼠气道炎症表型的影响,并分析其可能的原因。方法 BALB/c小鼠于实验第0、7、14、21、28、35天进行PBS液或HDM液(100μg/50μl PBS)滴鼻。采用头体积描记法测量小鼠的气道反应性;对支气管肺泡灌洗液(bronchoalveolar lavage fluid,BAL)进行细胞计数和分类;ELISA法检测BAL中CXCL1、CXCL2、CCL11(eotaxin)、IL-5、IFN-γ、IL-10和IL-17的含量及血清总Ig E;对肺组织PAS染色后进行病理学观察。结果 HDM1诱导了以中性粒细胞和嗜酸性粒细胞为主的混合型气道炎症;HDM2诱导的小鼠气道炎症以中性粒细胞升高为主;HDM3仅诱导了微弱的小鼠气道炎症反应,未见中性粒细胞和嗜酸性粒细胞的显著增加。3种HDM均引起BAL中CXCL1蛋白水平的升高,并伴随着高脚杯细胞增生等病理组织学改变。结论不同来源的HDM可能与不同类型的小鼠气道炎症相关。
Objective To investigate the effect of house dust mite extracts( HDM) on airway inflammation in a murine model. Methods BALB / c mice received an intranasal instillation of PBS or3 different HDMs( 100 μg) on the 0,7th,14 th,21th,28 th,35th days. After the final challenge,airway responsiveness( AR),histological changes in the airways and immunological status were examined. Results HDM1 induced the most robust influx of both neutrophils and eosinophils into the airways,HDM2 exerted a stronger effect on the induction of neutrophil airway inflammation,while HDM3 only elicited a very weak inflammatory response. Conclusion Different origins of HDM might contribute to different phenotypes of murine airway inflammation.
