Expression of Nerve Growth Factor and Proliferation Cell Nuclear Antigen in Glioma

摘要 目的:研究人脑胶质瘤中神经生长因子(NGF)和增殖细胞核抗原(PCNA)的表达,探讨它们与胶质瘤病理分级的关系及其在评估胶质瘤患者预后中的作用。方法:采用SP免疫组织化学法检测10例正常脑组织和64例胶质瘤中NGF和PCNA的表达,并分析其在正常脑组织及不同级别胶质瘤间的差异。结果:在正常脑组织与胶质瘤之间,以及高级别胶质瘤组与低级别胶质瘤组之间,NGF和PCNA的表达均存在显著差异(P<0.05),且随肿瘤病理级别的升高而增高;两者在生存时间>2年组与生存时间≤2年组之间的差别显著(P<0.05);相关性分析显示二者的表达存在正相关。结论:NGF和PCNA在胶质瘤的发生、发展过程中可能起重要的作用,其表达水平高低与胶质瘤的恶性程度及患者预后密切相关。 Objective:To examine the expression of nerve growth factor(NGF)and Proliferation cell nuclear antigen(PCNA)in human brain gliomas,and to investigate the effects of NGF and PCNA on prognosis of patients with glioma.Methods:The expressions of NGF and PCNA in 64 cases of glioma tissue and 10 cases of normal brain tissue were detected by immunohistochemical SP technique.Results:The positive expressions rates of NGF and PCNA in human gliomas of gradesⅠ-Ⅱ were significantly higher than those in normal brain tissues(P〈0.05),and significantly lower than those in human gliomas of gradesⅢ-Ⅳ(P〈0.05).The expression of NGF was positively correlated with the expression of PCNA.Conclusion:The level of NGF and PCNA in gliomas was upregulated higher than that of the normal controls.The results indicate NGF and PCNA may play an important role in the tumorigenesis and progression of gliomas,and relate to the extent of gliomas' malignant and patients' prognosis.
