摘要
目的:分析乳腺癌病灶组织解整合素样金属蛋白酶8(Adam8)和人类表皮生长因子受体2(Her2)表达的相关性,分析其在乳腺癌分型中的表达差异。方法:选择2012年2月至2014年1月期间来我院接受乳腺癌治疗的患者253例,按照乳腺癌TNM分期,分成T期组(n=115)、N期组(n=92)和M期组(n=46),每组纳入23例患者。采集病灶组织和癌旁组织,采用荧光定量PCR检测所有患者病灶组织和癌旁组织Adam8和Her2mRNA转录量,并计算病灶组织和癌旁组织转录量的比例,并比较不同组的比例。采用免疫组化检测所有患者病灶组织和癌旁组织Adam8和Her2阳性率,以及病灶组织和癌旁组织阳性率的比例,并比较不同组比例。结果:N期的Adam8和Her2mRNA转录量与癌旁组织转录量的比例为1.93±0.22和1.45±0.18,显著高于T期的1.48±0.11和0.98±0.08(t=17.91,P=0.000;t=23.27,P=0.000),M期的Adam8和Her2mRNA转录量与癌旁组织转录量的比例为2.84±0.33和2.02±0.35,显著高于N期(t=16.92,P=0.000;t=10.38,P=0.000)。N期的Adam8和Her2mRNA阳性率与癌旁组织阳性率的比例为2.93±0.53和1.93±0.23,显著高于T期的1.45±0.13和0.98±0.17(t=17.91,P=0.000;t=23.27,P=0.000),M期的Adam8和Her2阳性率与癌旁组织阳性率的比例为4.73±0.74和2.21±0.34,显著高于N期(t=16.92,P=0.000;t=10.38,P=0.000)。结论:对病灶Adam8和Her2表达量检测对于乳腺癌早期诊断以及疾病严重程度评价具有潜在的临床价值。
Objective:To analyze the correlation of a disintegrin and metalloproteinase 8(Adam8)and expression of human epidermal growth factor receptor 2(Her2)in breast cancer,to reveal their expression difference in the lesions of breast cancer.Methods:A total of 253 patients,who were diagnosed as breast cancer and received treatment during February 2012 to January 2014 in our hospital,were divided into T group(n=115),N group(n=92)and M group(n=46)according to the TNM staging method.Lesion tissues and paracancerous tissues were collected,fuorescence quantitative PCR was performed to measure Adam8 and Her2mRNA expression in lesion tissues and paracancerous tissues of all patients and tumor,and immunohistochemical assays were used to detect the positive rates of Adam8 and Her2.Results:The Adam8 and Her2mRNA expression in lesion tissues related to those in paracancerous tissues of N group was(1.93±0.22)and(1.45±0.18),which were significantly higher than those of T group(t=17.91,P=0.000;t=23.27,P=0.000),and the Adam8 and Her2mRNA expression in lesion tissues related to those in paracancerous tissues of M group was(2.84±0.33)and(2.02±0.35),which was significantly higher than those of N group(t=16.92,P=0.000;t=10.38,P=0.000).The positive rate of Adam8 and Her2in lesion tissues related to those in paracancerous tissues of N group was(2.93±0.53)and(1.93±0.23),which was significantly higher than those of T group(t=17.91,P=0.000;t=23.27,P=0.000),and the positive rate of Adam8 and Her2in lesion tissues related to those in paracancerous tissues of M group was(4.73±0.74)and(2.21±0.34),which was significantly higher than those of N group(t=16.92,P=0.000;t=10.38,P=0.000).Conclusion:Adam8and Her2 expression detection has a potential clinical value for the early diagnosis of breast cancer and disease severity evaluation.
