硫酸软骨素蛋白聚糖对MC3T3-E1细胞株矿化的影响

摘要 目的:观察硫酸软骨素蛋白聚糖(chondroitin sulfate proteoglycan,CSPG)对前成骨细胞株MC3T3-E1矿化过程中碱性磷酸酶(alkaline phosphatase,ALP)活性和钙离子沉积量的影响。方法:MC3T3-E1细胞株在含不同浓度CSPG(0、5、10、15mg/L)诱导液中培养,不加CSPG组为对照组,其余3组为实验组,细胞诱导14d时行ALP染色和活性检测,21d时行茜素红染色和钙离子沉积量检测。结果:与对照组比较,各实验组ALP和茜素红染色面积随CSPG浓度增加逐渐减小,颜色变浅;定量分析结果显示,钙离子沉积量整体差异有统计学意义(P<0.05),ALP活性整体差异无统计学意义。结论:在本实验周期内,CSPG以剂量依赖方式抑制MC3T3-E1细胞株钙离子的沉积。 Objective:To observe the effect of chondroitin sulfate proteoglycan(CSPG)on the alkaline phosphatase(ALP)activity and calcium deposition quantity in pre-osteoblast cell line MC3T3-E1 mineralization process.Methods:MC3T3-E1 cells were cultured with different concentrations of CSPG(0,5,10,15μg/ml)in induction medium,without CSPG as control group,the other three groups as the experimental group.Cells were cultured for 14 days by ALP staining and activity assay,21 days by alizarin red staining and calcium deposition assay.Results:Compared to the control group,ALP staining and alizarin red staining showed a decreasing trend with the increasing of CSPG concentration in the experimental group;The groups was statistically significant difference between the amount of calcium deposition(P〈0.05),no significant difference in ALP activity.Conclusion:In the experimental period,CSPG inhibited calcium deposition of MC3T3-E1 cells in a dose dependent manner.
