HPLC-ELSD法同时测定黄芪及金芪降糖片中5种皂苷类成分的含量

Simultaneous Determination of Five Astragalosides in Astragali Radix and Jinqi Jiangtang Tablet by HPLC-ELSD

目的：建立HPLC-ELSD法同时测定黄芪及金芪降糖片中黄芪甲苷、黄芪皂苷Ⅰ、黄芪皂苷Ⅱ、黄芪皂苷Ⅲ、异黄芪皂苷Ⅱ5种成分的含量。方法：色谱柱为Grace Apollo C18（250 mm×4.6 mm,5μm）;流动相为乙腈（A）-水（B）,梯度洗脱;体积流量：1.0 m L/min。结果：5种皂苷类成分线性关系良好,加样回收率在95%~105%。运用该方法测定了12批黄芪药材和10批金芪降糖片中5种皂苷类成分的含量。结论：该实验建立的方法专属性强,灵敏度高,简便易行,可用于同时测定黄芪及金芪降糖片中5种皂苷类成分的含量。

Objective： To develop an HPLC-ELSD method for simultaneous determination of Astragaloside Ⅳ,Astragaloside Ⅰ,Astragaloside Ⅱ,Astragaloside Ⅲ and Isostragaloside Ⅱ in Astragali Radix and Jinqi Jiangtang tablet. Methods： The chromatographic conditions were as follows： Grace Apollo C18column（250 mm × 4. 6 mm,5 μm）,acetonitrile（A） and water（B） as mobile phases for gradient elution,and the flow rate being 1. 0 m L/min. Results： Five components showed good linearity. The average recoveries were between95% ~ 105%. Five Astragalosides were determined in twelve batches of Astragali Radix and ten batches of Jinqi Jiangtang tablet. Conclusion： This is a specific,sensitive and simple method for simultaneous determination of Astragaloside Ⅳ,Astragaloside Ⅰ,Astragaloside Ⅱ,Astragaloside Ⅲ and Isostragaloside Ⅱ in Astragali Radix and Jinqi Jiangtang tablet.
