雪莲中绿原酸、芦丁、木犀草素含量的HPLC法测定

摘要 建立测定雪莲(Saussurea involucrata)中绿原酸、芦丁和木犀草素含量的方法,采用HPLC法,色谱柱为Agilent Eclipse XDB-C18(250 mm×4.6 mm,5μm),乙腈-0.8%磷酸水溶液进行梯度洗脱,流速1.0m L/min,检测波长360 nm,柱温30℃。绿原酸、芦丁、木犀草素的线性范围分别为0.081-0.408μg/m L、0.090-0.449μg/m L、0.045-0.224μg/m L;平均加样回收率分别为99.78%、99.89%、100.70%,其中绿原酸、芦丁在天山雪莲中含量最高,木犀草素在苞叶雪莲中含量最高。 To establish a method for determining chlorogenic acid, rutin and luteolin in Saussurea involucrate, the Agilent Eclipse XDB-C18(250 mm ×4.6 mm, 5μm) column was used. The mobile phase was consisted of acetonitrile and 0.8%aqueous phosphoric acid with gradient elution.The flow rate was 1. 0 ml·min-1 and the detective wavelength was 360 nm.The column temperature was 30℃. The peak areas and concentrations of chlorogenic acid, rutin and luteolin had good correlation in the ranges of 0.081 -0.408μg / m L,0.090 -0.449μg / m L and 0. 0448 - 0.224μg / m L, with the average recovery rates of 99.78%,99.89% and 100.70%, respectively. The contents of chlorogenic acid and rutin were the highest in Saussurea involucrata(Kar. et Kir.) Sch.-Bip. Saussurea obov-allata(DC.) Edgew had the maximum content of luteolin.
