摘要
目的探讨CD147蛋白和凋亡抑制基因Survivin在膀胱癌中的表达及其相互关系。方法收集2012年1月至2014年6月在我院手术治疗的42例膀胱癌患者的癌组织和24例前列腺增生手术所获得的正常膀胱组织,RT-PCR方法检测膀胱癌组织和正常膀胱组织中Survivin mRNA的表达,免疫组化SP法检测CD147蛋白在膀胱癌和正常膀胱组织中的表达,对相关结果进行分析。结果正常膀胱组织中无Survivin mRNA表达,膀胱癌组织中Survivin mRNA表达阳性率为76.19%,CD147蛋白在正常膀胱组织中的表达阳性率为4.17%,在膀胱癌组织中为78.57%。42例膀胱癌患者中CD147蛋白和Survivin mRNA均表达阳性为31例,双阳性表达率为73.81%,双阴性表达率为19.04%。Spearman相关分析显示CD147蛋白和Survivin mRNA在膀胱癌中的表达呈正相关(r=0.798,P<0.05);CD147蛋白、Survivin基因在膀胱癌组织中的表达与TNM分期呈正相关。结论 CD147蛋白和凋亡抑制基因Survivin在膀胱癌组织中有较高表达,Survivin基因的异常表达与CD147蛋白的上调可能在膀胱癌的发展中起协同作用。
Objective To investigate the expression and relationship of CD147 protein and apoptosis inhibiting gene Survivin in bladder cancer. Methods CD147 protein and apoptosis inhibiting gene Survivin were detected in 42 cases of bladder cancer tissues and 24 cases normal bladder tissues,from 2012 January to 2014 June in our hospital operation treatment,RT-PCR was used to detect the expression of Survivin mRNA in bladder cancer tissue and normal bladder tissue,immunohistochemical SP method was used to detect the expression of CD147 protein in bladder carcinoma and normal bladder tissues,the results were analysed statistically. Results No Survivin gene expression in normal bladder tissue,expression rate of Survivin gene in bladder cancer was 76.19%,the positive rate of CD147 expression was 4.17%in the normal bladder tissue,the positive rate of CD147 expression was 78.57%in bladder cancer tissues;42cases of patients with bladder cancer CD147 protein and Survivin gene expression were positive in 31 cases,double positive expression rate was 73.81%,double negative expression rate of 19.04%.The Spearman correlation analysis,significant positive correlation between CD147 protein and Survivin gene in bladder cancer(r=0.798,P〈0.05);CD147protein and Survivin gene expression in bladder cancer tissue positively correlated with the TNM staging(P〈0.05).Conclusions Protein CD147 and apoptosis suppressor gene Survivin has higher expression of its expression is closely related to both,abnormal expression of Survivin gene and protein CD147 upregulation may play a synergistic role in the development of bladder cancer.
