摘要
SRAP是一种可检测基因可译框区域多态性的分子标记系统,具有简便、稳定、易测序等优点。本研究利用SRAP标记对16份浙江柚类地方资源及2份柚与其他柑橘的杂交后代进行了遗传多样性研究。结果显示,49对引物中筛选出13对多态性引物,引物组合扩增条带数在2~14条,平均为9条带,多态率为0~100%,平均为73.5%。种质间的相似系数从0.513到0.932,其中四季柚和红心四季柚的相似系数最高,而管南土柚和玉环柚为最低。聚类分析结果显示,样品在遗传相似系数0.738处分为四类,第一类包括13份样品,第二类包括管南红肉抛和管南土柚,第三类为皋泄香柚,第四类包括常山胡柚和槾文柑。研究结果表明,浙江柚类资源遗传背景复杂,大多数种质由外地引入并驯化而成。
Sequence related amplified polymorphism(SRAP) was kind of molecular marker systems with the advantages of simplicity, stability and ready to sequence, and could detect polymorphism of open reading frames.SRAP markers were employed for identification and genetic diversity analysis of 16 native pummelo(C. grandis Osbeck) germplasms from Zhejiang province and 2 hybrids from the cross of pummelo and other citrus in the study.The results showed that 13 SRAP primer combinations generating distinct bands were chosen for cultivar identification from 49 primer combinations. The number of bands by primer combinations ranged from 2 to 14,with an average of 9 bands. The number of polymorphic bands ranged from 0~100% with an average of 73.5%.The genetic similarity coefficients between different gemplasms ranged from 0.513 to 0.932. From that, the value between Siji pummelo and Hongxinsiji pummelo showed the highest,while that between Guannan pummelo and Yuhuan pummelo were the lowest. UPGMA cluster analysis based on the SRAP fragments showed that the 18 gemplasms could be divided into four major groups on similarity coefficient level of 0.738, among which 13 varieties were clustered in the first group, Guannanhongrou pummelo and Guannan pummelo were clustered in the second group, Gaoxiexiang pummelo was clustered in third group, and Changshanhuyou and Manwengan were clustered in the fourth group. The results showed that pummelo germplasms from Zhejiang province had complex genetic background, and most of them were introduced from outland and domesticated in the local.
