健脾益智胶囊对MCAO大鼠海马β-catenin表达的影响 被引量:5

摘要 目的 :探讨健脾益智胶囊对大脑中动脉栓塞(MCAO)大鼠海马β-连环蛋白(β-catenin)基因与蛋白表达的影响。方法:SD大鼠120只,建立MCAO大鼠模型,随机分为正常组、假手术组、模型组、西药组、中药组,模型组、假手术组给予0.9%氯化钠溶液,中药组、西药组分别给予健脾益智胶囊、尼莫地平混悬液灌胃。采用蛋白免疫印迹、实时荧光定量PCR法于术后7 d、14 d、28 d观察海马β-catenin蛋白及m RNA表达情况。结果:术后7 d,中药组β-catenin蛋白及m RNA表达均高于其他各组(P<0.05);术后14 d中药组蛋白及m RNA表达高于正常组、假手术组、模型组(P<0.05);术后28 d中药组β-catenin蛋白及m RNA表达高于模型组(P<0.05),低于中药组7 d、14 dβ-catenin蛋白及m RNA表达(P<0.05)。结论:健脾益智胶囊可提高β-catenin蛋白及基因的表达,提示健脾益智胶囊能可能通过激活Wnt通路以促进缺血后神经干细胞增殖分化。 Objective:To explore and discuss the effects of Jianpi Yizhi capsule on gene and protein expression of β-catenin in the hippocampus of MCAO rats. Methods:120 SD rats model of MCAO were randomly divided into 5 groups:normal control group,sham operation group,model group,western medicine group and Chinese medicine group. The sham operation group was give 0.9% sodium chloride solution and the Chinese medicine group and westem medicine group were given Jianpi Yizi capsule and nimodipine mixture by gavage respectively. 7,14 and 28 days after the operation,the protein and m RNA levels of β-catenin(Notch1and Jagged1) in hippocampus were assayed respectively by Western blotting and Real-time PCR methods.Results:7 days after the operation,the protein and m RNA expression levels of β-catenin of the Chinese medicine group were significantly higher than all the other groups(P 〈0.05). 14 days after the operation,the protein and m RNA expression levels of β-catenin of the Chinese medicine group were higher than nor mal control group,sham operation group and model group(P 〈0.05). 28 days after the operation,the protein and m RNA levels of β-catenin of the Chinese medicine group were higher than model group,but lower than that on the 7th day and the 14 th day(P 〈0.05). Conclusion:Jianpi Yizhi capsule can increase protein and gene expression level of β-catenin,which suggests that the Jianpi Yizhi capsule may activate the Wnt pathway to regulate the the proliferation and differentiation of neural stem cells in the dentate gyrus of hippocampus after ischemia.
