摘要
目的探讨在神经细胞早期生长发育过程中Nogo-A与Nogo-A受体(NG-R)的表达及意义。方法体外培养PC12细胞,实验组以神经生长因子(NGF)诱导其分化,对照组不进行诱导。倒置显微镜下选取20个随机视野进行观察,计数轴突生长和细胞增殖情况。采用免疫荧光染色、逆转录酶聚合酶链反应(RT-PCR)等方法检测PC12细胞生长及分化过程中Nogo-A与NG-R m RNA及蛋白的表达与变化。结果对照组PC12细胞未检出Nogo-A与NG-R的表达。在实验组NGF诱导PC12细胞向交感神经细胞分化过程中,Nogo-A与NG-R表达量逐渐增高(P<0.05)。结论在神经细胞发育早期,Nogo-A与NG-R可能发挥促进神经轴突生长的作用。
Objective To explore the expression and its significance of Nogo-A and Nogo-A receptor(NG-R) in archineuron cells during differentiation and growth. Methods PC12 cells were cultured in vitro and induced by nerve growth factor(NGF) in experimental group.The axon growth was observed by inverted microscopy. The expressions of Nogo-A and NG-R m RNA and protein in PC12 cells were detected by semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR) and immunofluorescence staining during cell growth and differentiation. Results Nogo-A and NG-R expression was not detected in PC12 cells in the control group. When PC 12 cells were induced by NGF and differentiated to sympathetic nerve cells in the experimental group, the expressions of Nogo-A and NG-R increased gradually(P〈0.05). Conclusions During the early development of neuron, Nogo-A and NG-R may facilitate neurite growth.
