摘要
目的:建立一种测定磷酸奥司他韦胶囊含量的方法。方法采用高线液相色谱法测定磷酸奥司他韦的含量,色谱柱:Kromasil C8(250mm×4.6mm,5μm),流动相为0.05mol/L磷酸二氢钾溶液(40%氢氧化钾调节pH至5.5)-甲醇-乙腈(70:20:10),检测波长为207nm,柱温30℃,流速1.0mL/min,进样量20μL。结果磷酸奥司他韦在1~50μg范围内线性关系良好(r=0.9992)。平均回收率为99.8%,RSD=0.9%(n=9),稳定性、精密度、重复性试验的相对标准偏差均小于0.8%。结论此方法操作简便快捷、准确度高,适用于磷酸奥司他韦胶囊的含量测定。
ObjectiveTo establish an method for the determination of Oseltamivir Phosphate capsules.Methods HPLC method was adopted.The separation was performed on Kromasil C8(250mm×4.6mm,5μm), column with mobile phase consisted of 0.05mol/L postassiam dihydrogen phosphate (adjust pH to 5.5 with 40% potassium hydroxide)- methanol- acetonitrile(70:20:10). UV detection wavelength was set at 207nm,the column temperature was maintained at 30℃,the flow rate was 1.0mL/min and the sample size was 20μL.Results The line arrange of Oseltamivir Phosphate was 1-50μg,the correlation coefficient was 0.9990 and the average recovery was 99.8%(RSD=0.9%,n=9).RSDs of stability test,precision test,reproducibility test were all less than 0.8%. Conclusion The method is simple,quick, accurate and suitable for the content determination of Oseltamivir Phosphate capsules.
