摘要
目的探讨绞股蓝总苷对大鼠酒精性脂肪肝(AFL)的保护作用及机制。方法采用高脂饮食联合酒精灌胃6周建立大鼠AFL模型,药物干预组分别给予绞股蓝总苷及水飞蓟素200 mg/kg灌胃。采用生化法检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)的活性和肝匀浆超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GPx)活性、丙二醛(MDA)含量;HE染色光镜下观察肝组织病理学变化;免疫组化法检测肝脏核转录因子-κB(NF-κB)和肿瘤坏死因子-α(TNF-α)。结果绞股蓝总苷组血清ALT、AST水平明显低于模型组(P<0.05);绞股蓝总苷组肝匀浆MDA水平较模型组明显下降(P<0.05),SOD及GPx活性较模型组明显升高(P<0.05);光镜下显示绞股蓝总苷组肝脏脂肪变性明显减轻;肝组织NF-κB和TNF-α表达明显减少(P<0.05)。结论绞股蓝总苷对AFL所致的肝损伤具有一定的保护作用,其保护作用可能与抑制NF-κB、TNF-α的表达、抑制氧化应激,减少炎症因子的释放有关。
Objective To investigate the protective effects of Gynostemma total glucoside on alco-holic fatty liver in rats.Methods Rat model of alcoholic fatty liver was induced by intragastric in-fusion of ethanol and high-fat diet for six weeks.The drug intervention group was induced by intragastric infusion and was given Gynostemma total glucoside and silymarin 200 mg/kg at the same time.The activities of serum alanine aminotransferase, aspartate aminotransferase were measured by routine laborato ry methods using an autoanalyzer.The activities of superoixde dis-mutase (SOD) and glutathione peroxidase(GPx) and th e level of malondialdehyde (MDA) in liver group Gynostemma total glucoside could ameliorate ethanol-induced hepatic fat degenera-tion.Results The activities of serum ALT, AST inGynostemma total glucoside treatment group were markedly l ower than those in model group.Compared with model group,Gnyostemma total glucoside notablely decreased MDA content a nd increased GPx activity, and Gynostemma total glucoside remarkably increased SOD activity.Moreover, the levels of NF -κB and TNF-αex-pression in rat liver Gynostemma total glucoside treatment group was significantly lower than those in model group.Conclusion Gynostemma total glucoside had a certain protective effect in al-coholic fatty liver and protective effects on alcoholic fatty liver may be related to inhibiting the expression of NF-κB and alleviating oxidative stress.
