GC-MS法分析比较两种处理方式所得酵母细胞自溶成分

摘要 为分析添加不同促溶剂(Na Cl和木瓜蛋白酶)所得酵母细胞自溶物的成分差别,采用气相色谱-质谱联用(GC-MS)技术,对酵母细胞自溶物乙酸乙酯萃取物中的化学成分进行分析测定,结合计算机检索技术对其成分进行结构鉴定,应用色谱峰面积归一化法计算各成分的相对百分含量。结果表明,添加Na Cl所得自溶物的乙酸乙酯萃取物共分离出49种成分,鉴定出28种,占总量的92.14%,其中酯类3种,酸类5种,醇类4种,杂环类6种(吡咯、呋喃、吡咯并吡嗪、嘧啶、吡唑等),酚类2种,饱和芳香烃3种,饱和脂肪烃5种;添加木瓜蛋白酶所得自溶物的乙酸乙酯萃取物中共分离出49种成分,鉴定出27种,占总量的91.006%,其中酯类2种,酸类4种,醇类4种,杂环类7种(吡咯、呋喃、吡咯并吡嗪、嘧啶、噻唑、吡喃等),酚类5种,饱和芳香烃2种,不饱和芳香烃2种,腈类1种。由此可见,不同处理方式所得酵母细胞自溶物乙酸乙酯萃取物共有21种成分,其余成分需进一步研究。 In order to analyze the differences of material composition in the cells yeasted by the adding different au- tolyzing solvent (NaC1 and papain),the gas chromatography - mass spectrometry (GC-MS) technology were used to ana- lyze and examine the chemical composition of autolyzing solvent,and the structural identification was conducted by using computer retrieval techniques and the relative percentage of each component was calculated with chromatographic peak area normalization method.The results showed that 49 components were isolated from ethyl acetate extracts added NaCI and 28 components were identified,accounting to 92.14% of the total,in which there were three kinds of esters ,five kinds of acids,four kinds of alcohol,six kinds miscellaneous rings (pyrrole,furan,pyrrole and pyrazine,pyrimidine,pyrazole, etc.) ,two kinds of phenols,three kinds of unsaturated aromatic hydrocarbons,five kinds of saturated aliphatic hydrocar- bons.The results showed that 49 components were isolated from ethyl acetate extracts added papaya protease and 27 components were identified, accounting to 91.006% of the total,of which there were two kinds of esters,four kinds of acids,four kinds of alcohols, seven kinds of heterocyclic (pyrrole,furan,pyrrole and pyrazine,pyrimidine,thiazole,pyran, etc.) ,five kinds of phenols,two kinds of saturated hydrocarbons,two kinds of the unsaturated aromatic hydrocarbon and one kind of nitriles.The results indicated that:the ethyl acetate extracts from the yeast cells through different treatment in- clude a total of 21 kinds of ingredients and the remaining ingredients need further study.
