摘要
目的探讨黄芪桂枝五物汤中总皂苷对机体免疫功能的影响,从而为相关产品开发和临床应用提供实验依据。方法取40只小鼠,随机分为皂苷组1(低剂量),皂苷组2(中等剂量)和皂苷组3(高剂量)以及正常对照组,每组10只。总皂苷组每天灌服0.2ml浓度为5,10,20mg/ml的总皂苷溶液,正常对照组每天灌服0.2ml生理盐水,10d后处死,取样测定小鼠各项免疫指标。1检测小鼠巨噬细胞吞噬功能;2小鼠后足迟发型超敏反应强度;3血凝法测定小鼠血清中溶血素含量;4对脾脏和胸腺称重计算脾脏指数和胸腺指数。结果皂苷各组小鼠巨噬细胞吞噬率和吞噬指数明显低于对照组(P<0.01);皂苷各组小鼠左右后足重量差与对照组小鼠相比,有明显降低(P<0.01);皂苷各组小鼠抗体积数低于对照组(P<0.05);皂苷各组小鼠脾脏指数和胸腺指数均较对照组小鼠有明显降低(P<0.01)。结论黄芪桂枝五物汤中总皂苷可以显著降低机体的部分固有免疫功能、细胞免疫功能和体液免疫功能。
Objective To explore the effects of saponin in Huangqi Guizhi Wuwu on immune function for the clinical application and industrial production.Methods Forty mice were randomly divided into four groups,10 mice in each group:saponin group 1(low dose),saponin group 2(middle dose),saponin group3(high dose)and normal control(NC)group.Saponin groups were fed with 0.2ml of saponin(5 mg/ml,10 mg/ml,20 mg/ml respectively)per day,and NC group 0.2ml of NS.After 10 day feeding,the mice were killed without pain and the samples were collected from them.The immune function of the mice was examined as follows.(1)The macrophage function was detected;(2)Weight difference of left and right postpedes were checked;(3)Antibody aggregate was studied;(4)Spleen and thymus indexes were researched.Results Phagocyte rate and phagocyte index of saponin groups were significantly lower than that of the NC group(P〈0.01).Compared with NC group,weight difference of left and right postpedes of saponin groups were significantly improved.Antibody aggregate of saponin groups was also decreased(P〈0.05).Compared with NC group,spleen and thymus index of saponin groups were significantly decreased.Conclusion Saponin can decrease parts of immune functions with innate immune,humoral immune and cellular immune.
