摘要
利用海藻酸钠为载体包埋制备固定化α-淀粉酶,在海藻酸钠浓度,氯化钙浓度和游离酶添加量的单因素实验基础上,采用响应曲面设计对三因素进行优化确定固定化的最优条件。得到的最佳条件为:海藻酸钠浓度为2.48%、氯化钙浓度为2.04%、游离酶浓度为0.23%,在该条件下进行验证实验得到固定化酶的回收率为74%,达到了较高的固定化酶的回收率。
α-amylase was immobilized using sodium alginate by a embedding-crosslinking method.On the base of single factor experiment, the optimal conditions were determined by using response surface methodology.The results showed that the optimal conditions were 2.48% sodium alginate, 2.04% CaCl2 and 0.23% α-amylase, the high immobilization efficiency (74%) was obtained in the optimal conditions.
