摘要 目的研究虫草素对高转移潜能乳腺癌细胞株MDA-MB-231细胞增殖、凋亡及其TMSG-1蛋白表达的影响。方法采用MTT法观察不同浓度的虫草素(0、1、2、4和8μg/ml)对MDA-MB-231细胞增殖的作用;流式细胞仪检测细胞凋亡率变化;免疫印迹法检测经不同浓度的虫草素(0、1、2、4和8μg/ml)处理48 h后,MDA-MB-231细胞TMSG-1蛋白的表达。结果虫草素能够以剂量依赖方式显著地抑制MDA-MB-231细胞增殖(P<0.01)。流式细胞术结果显示,虫草素可显著促进MDA-MB-231细胞凋亡(P<0.01)。虫草素可以呈剂量依赖方式增强MDA-MB-231细胞TMSG-1的表达。与对照组相比,2μg/ml虫草素即可明显上调MDAMB-231细胞TMSG-1蛋白的表达(P<0.05)。结论虫草素可以呈剂量依赖方式抑制MDA-MB-231细胞的生长并诱导其凋亡;虫草素可能通过上调MDA-MB-231细胞TMSG-1蛋白的表达,发挥其潜在的抑制肿瘤细胞浸润转移的作用。 [Objective] To investigate the effect of cordycepin on cell proliferation and apoptosis and TMSG-1 protein expression in high metastatic potential breast cancer cell line MDA-MB-231 cells. [Methods] The MDA-MB-231 cells were incubated with cordycepin solution (0, 1, 2, 4 and 8 μg/ml, respectively) for different time (12, 24 and 48 h, respectively). Effect of cordycepin solution on MDA-MB-231 cell proliferation was detected by MTT assay. Cell apoptosis rates were determined by flow cytometry detection. The effect of cordycepin on the expression of TMSG-1 protein in the MDA-MB-231 cell lines was detected by Western blot. [Results] Cordycepin significantly inhibited the proliferation of MDA-MB-231 cells in a dose dependent manner(P〈0.01). Cordycepin significantly promoted the apoptosis of MDA-MB-231 cells in a dose and timedependent manner (P〈0.01). The expression of TMSG-1 protein was increased by cordycepin in a dose-dependent manner in MDA-MB-231 ceils. The expression of TMSG-1 protein was significantly upregulated by cordycepin (〉 2μg/ml) in MDA-MB-231 cells compared with the untreated cells (P〈 0.05). [Conclusions] Cordycepin could significantly suppress MDA-MB-231 cell proliferation and induce apoptosis in a concentration-dependent manner, and upregulate the expression of TMSG-1 protein in the cells. It is suggested that cordycepin maybe play a potential role in inhibiting tumor cell invasion and metastasis.
