摘要 目的检测FOXO1在胃正常组织与胃癌组织中的表达水平,探讨其与胃癌临床病理因素的关系。方法采用免疫组织化学、实时荧光定量PCR法对收集到的28例因胃良性溃疡病变切除的标本以及32例胃癌切除标本中FOXO1表达情况进行检测,并分析其与胃癌之间的关系。结果胃良性溃疡组正常胃组织内的FOXO1 mRNA及蛋白都呈低表达,而胃癌组织内的FOXO1 mRNA及蛋白表达升高(P<0.05);高分化胃癌组织中FOXO1 mRNA及蛋白表达显著高于中低分化胃癌组织(P<0.05),胃癌组织中FOXO1 mRNA及蛋白表达与患者性别、年龄、肿瘤部位、TNM分期及淋巴结有无转移无明显相关(P>0.05)。结论 FOXO1在胃正常组织和胃癌组织中的表达存在差异,其表达水平与胃癌分化程度相关,可能在胃癌的发生发展过程中起重要作用。 [Objective] To investigate the expressions of FOXO1 in normal gastric tissue and gastric carcinoma, and to explore its relationship with the clinical pathologic factors of gastric cancer. [Methods] The expressions of FOXO1 in normal gastric tissue of 28 patients with gastric ulcer and gastric cancer tissues of 32 patients with gastric cancer were detected by immunohistochemical staining and real-time quantitative PCR, and its relationship with gastric cancer was analyzed. [Results] The expressions of FOXO1 mRNA and protein in the normal gastric tissue of the benign gastric ulcer group were both low, while their expressions in the gastric cancer tissues increased (P〈 0.05). The expressions of FOXO1 mRNA and protein in the well-differentiated gastric cancer tissues were significantly higher than those in the poorly-differentiated gastric cancer tissues (P〈 0.05). There was no significant correlation between the expressions of FOXO1 mRNA and protein in the gastric cancer tissues and gender, age, tumor location, TNM stage and lymph node metastasis of the pa- tients (P〉 0.05). [Conclusion] FOXO1 shows different expression levels in the normal tissue of benign gastric ulcer and gastric cancer tissues. FOXO1 expression level is correlated with the pathological differentiation of gastric cancer, which may play a role in the occurrence and development of gastric cancer.
