骨髓间充质干细胞对再生障碍性贫血患者FoxP3基因表达的影响研究 被引量:2

摘要 目的研究骨髓间充质干细胞(MSC)对再生障碍性贫血(AA)患者外周血Fox P3m RNA表达量的影响,并初步探讨MSC发挥作用的机制。方法分离AA患者外周血T淋巴细胞,与体外培养扩增的健康供者MSC按3:1比例行共培养,共培养方式分为直接接触法及Transwell法,共培养72 h后收集T淋巴细胞,提取细胞总RNA,逆转录成c DNA,实时定量PCR检测共培养前后Fox P3m RNA的相对表达量。结果 T淋巴细胞与MSC共培养72 h后Fox P3m RNA相对表达量较前升高,直接接触组为9.13,Transwell组为5.18。结论 MSC可通过与T细胞直接接触或旁分泌机制上调AA患者外周血T细胞的Fox P3m RNA表达。 Objective To study the effects of mesenchymal stem cells (MSC) on FoxP3 gene expression in T cells from patients with aplastic anemia and to explore its possible mechanism. Methods T lymphocytes were isolated from the peripheral blood of patients with AA. MSC were co-cultured with T lymphocytes at 3:1 ratio. Additionally, MSC were separated from T lymphocytes with a Transwell system and cultured at 3:1 ratio. After 72 hours, T lymphocytes were collected and total cellular RNA were extracted for reverse transcription. Relative expression of FoxP3 mRNA was detected through real-time quantitative PCR. Results After co-culture with MSC in a well for 72 hours, the relative expression of FoxP3 mRNA increased 8.13 folds. When co-cultured with MSC in a Transwell system, the relative expression of FoxP3 mRNA increased 4.18 folds. Conclusions MSC can up-regulate FoxP3 mRNA expression in T cells from patients with AA through both direct contact and secretion of cytokines.
