摘要
为探明核转录因子kappaB(NF-κB)抑制剂吡咯烷二硫代甲酸铵(PDTC)对兔肾下腹主动脉瘤(AAA)形成的抑制作用,以及为临床开发和应用相关药物治疗AAA提供理论依据,对15只新西兰大耳兔用0.75mol/L CaCl2溶液浸润肾下腹主动脉制成AAA模型,随机平分为3组,即模型对照组、吡咯烷二硫代甲酸铵(PDTC)组、生理盐水组,用ELISA法测定各组血清基质金属蛋白酶-9(MMP-9)、白细胞介素6(IL-6)、兔肿瘤坏死因子α(TNF-α)含量,用游标卡尺测定AAA直径。结果表明:PDTC组每天灌服PDTC20mg/kg体重1mL,4周后AAA瘤体直径较模型对照组和生理盐水组极显著减小(P<0.01),与用药前相比无显著性增大;PDTC组循环促炎性因子IL-6、TNF-α含量极显著降低(P<0.01),MMP-9含量显著降低(P<0.05)。PDTC作为炎性因子枢纽NF-κB的抑制剂能显著抑制炎性反应,从而抑制AAA的形成。
The abdominal aortic aneurysms models of 15 New Zealand white rabbits infiltrated with 0.75 mol/L CaCl2 were successfully built,then 15 AAA models were divided into three groups (CK, PDTC and normal saline groups)randomly and finally the MMP-9,IL-6 and TNF-αcontent and AAA diameter of CK,PDTC and normal saline groups were determined by ELISA to study the inhibition effect of PDTC on rabbit&#39;s abdominal aortic aneurysms and to provide the theoretical basis for clinic development and application of relative drugs to cure AAA.The results showed that the AAA diameter of PDTC group fed with 20mg PDTC per kg of body weight for each day after 4 weeks is smaller than CK and NS groups very significantly and there is no obvious difference in AAA diameter before and after PDTC use in PDTC group.The IL-6 and TNF-α content and MMP-9 content of PDTC group after 4 weeks decrease very significantly and significantly compared with IL-6 and TNF-αcontent and MMP-9 content before PDTC use respectively.PDTC can inhibit inflammatory reaction significantly,which inhibits AAA formation.
