国内活禽市场新型环形病毒AGV2的发现 被引量:1

Detection of a Novel Gyrovirus AGV2 in Live Poultry Market in China's Mainland

摘要 为了解国内鸡群是否存在新型环形病毒AGV2,研究对来自扬州的一个活禽市场的20份鸡羽囊样品进行了AGV2的PCR检测。通过对鸡羽囊DNA提取、PCR扩增以及序列分析,在20份样品中检测到4份阳性样品。测序比对发现,本研究检测到的国内AGV2序列与NCBI中发表的AGV2的同源性为95.7%~99.4%。并对一份阳性样品的PCR产物进行了TA克隆,测定了该PCR的灵敏度为2.7个拷贝数。这一结果不仅首次证明国内活禽市场存在AGV2,而且为进一步开展国内鸡群AGV2的分子流行病学及其致病性等研究奠定良好的基础。 To investigate whether the novel gyrovirus AGV2 is present in chicken flock in China's Mainland,using PCR detection for AGV2 in chickens from a live poultry market in Yangzhou was performed. Through DNA extraction,PCR amplification and sequencing,4 in 20 samples of chicken feather shafts were positive for AGV2. Sequence analysis revealed that these four AGV2 sequences identified here showed 95.7% to 99.4% homology to those deposited in NCBI database. In addition,TA-cloning was conducted for one positive sample,and it was found that the limit detection of the PCR for AGV2 was 2.7 copies. Our data not only demonstrated the detection of AGV2 in chickens in China's Mainland for the first time,but also pave the way for further investigating the molecular epidemiology and pathogenesis of AGV2 in China.
