摘要
为明确白细胞介素8基因(IL8)在浙江省内不同牛种群体间的遗传变异情况,应用PCR-SSCP技术检测了中国荷斯坦牛、温岭高峰牛和温州水牛IL8基因3'UTR的多态性。PCR-SSCP和测序结果表明,中国荷斯坦牛和温岭高峰牛在IL8基因3'非翻译区存在3个完全连锁的SNP(g.2831A/G,g.2904 T/C,g.3030 G/A)构成的单倍域,温州水牛在这些位点均表现为突变纯合型。利用一般线性模型分析IL8基因3'UTR的3个SNPs对中国荷斯坦牛305 d校正产奶量、乳脂量、乳蛋白率及体细胞评分的影响,发现这些SNPs可显著影响中国荷斯坦牛305 d校正产奶量(P<0.01)和乳脂率(P<0.05)。
In order to clarify interleukin 8(IL8) gene in the genetic variability of different domestic cattle populations,we detected IL8 gene 3' untranslated region mutations among Chinese Holstein,Wenling humped cattle and Wenzhou buffalo by application of PCR-SSCP technique. One haplotype block,containing g. 2831 A / G,g. 2904 T /C,and g. 3030 G / A was identified in 3' UTR of IL8 gene by PCR-SSCP and DNA sequencing in Chinese Holstein and Wenling humped cattle,while Wenzhou buffalo individuals were homozygous for the "mutant allele"in the same loci. The effect of IL8 gene 3' UTR polymorphisms on Chinese Holstein 305-day corrected milk yield fat percentage,protein percentage and somatic cell score was assessed by general liner mode. Three mutations in IL8 gene 3' UTR were significantly associated with Chinese Holstein 305-day corrected milk yield(P 0. 01) and fat percentage(P 0. 05).
