桂竹糖芥苷对大鼠肾髓袢升支粗段管周膜50 pS钾通道活性的调控机制 被引量:1

摘要 目的:探讨桂竹糖芥苷对肾髓袢升支粗段(thick ascending limb,TAL)管周膜50 p S钾通道活性的调控机制。方法:采用细胞贴附式膜片钳技术及免疫印迹技术测定桂竹糖芥苷对50 p S钾通道活性的调控机制。结果:桂竹糖芥苷可明显抑制50 p S钾通道的活性(P<0.01);PKC阻断剂存在时,桂竹糖芥苷不再增加50 p S钾通道的活性(P>0.05)。结论:桂竹糖芥苷可能是通过激活PKC依赖途径抑制50 p S钾通道活性的。 Objective: To study the regulatory mechanism of erysimosides on the 50 p S potassium channel in the thick ascending limb( TAL) of rat kidney. Methods: The patch- clamp cell- attached recording technique was used to record the single potassium channel current. The western blot technique was used to detect the protein express of phosphate- PKC. Results: Erysimosides can decrease the 50 p S potassium channel activity of TAL,and PKC inhibitor can inhibit the effect. Conclusion: Erysimosides can inhibit the activity of basolateral 50 p S potassium channel by the PKC- dependent pathway.
