磷脂酶A2-ⅡA亚型基因rs11677 C/T多态位点基因突变及蛋白表达与食管鳞状细胞癌临床病理特征的关联性研究 被引量:2

摘要 目的探讨磷脂酶A2-ⅡA亚型(PLA2G2A)基因中的单核苷酸多态性(SNP)rs11677 C/T多态位点基因突变及蛋白表达与食管鳞状细胞癌(ESCC)临床病理特征的关系。方法选取2009年10月—2010年9月河南省林州市食管癌高发区,首次经组织病理学确诊的食管ESCC患者260例,血液标本采用Mass Array时间飞行质谱生物芯片检测PLA2G2A基因rs11677 C/T位点基因型;采用免疫组织化学染色方法 (SP法)检测癌及癌旁组织中PLA2G2A蛋白的表达情况。比较不同临床特征食管ESCC患者PLA2G2A基因rs11677 C/T阳性率及PLA2G2A蛋白表达阳性率,不同rs11677 C/T基因型食管ESCC患者PLA2G2A蛋白表达阳性率。结果 260例食管ESCC患者中PLA2G2A基因rs11677 C/T多态位点CC型66.2%(172/260)、CT+TT型33.8%(88/260)。携带CC基因型者PLA2G2A蛋白表达阳性率(56.4%,97/172)与携带CT+TT基因型者(55.7%,49/88)比较,差异无统计学意义(P>0.05)。不同年龄、组织学分化、TNM分期、淋巴结转移、浸润深度食管ESCC患者PLA2G2A基因rs11677 C/T基因型频率比较,差异均无统计学意义(P>0.05)。不同年龄、组织学分化、TNM分期、淋巴结转移食管ESCC患者PLA2G2A蛋白表达阳性率比较,差异均无统计学意义(P>0.05);不同浸润深度食管ESCC患者PLA2G2A蛋白表达阳性率比较,差异有统计学意义(P<0.05)。不同rs11677C/T基因型食管ESCC患者PLA2G2A蛋白表达阳性率比较,差异无统计学意义(χ2=0.149,P=0.928)。结论未发现PLA2G2A基因rs11677 C/T多态位点基因突变与其蛋白表达的相关性,PLA2G2A蛋白表达与食管ESCC浸润深度相关。 Objective To investigate the correlation among the phospholipase A2 group ⅡA( PLA2G2A) gene single nucleotide polymorphism( SNP)( rs11677 C / T),the protein expression and the clinicopathologic characteristics of esophageal squamous cell carcinoma( ESCC). Methods A total of 260 ESCC patients from Linzhou city,Henan province,an area of high incidence of esophageal cancer,were selected as study subjects from October 2009 to September 2010,ESCC was confirmedby histopathologic examination for the first time. Blood specimens were used to detect the PLA2G2 A rs11677 C / T SNP genotype by time-of-flight mass spectrometry Mass Array system, and the carcinoma and pericarcinous tissues were used to examine the expression of PLA2G2 A protein by immunohistochemical SP method. The positive rates of PLA2G2 A rs11677 C / T and PLA2G2 A protein expression were compared among different groups of ESCC patients with different clinical features,the positive rates of PLA2G2 A protein expression were compared among different groups of ESCC patients with different rs11677 C / T genotypes. Results Among 260 ESCC patients,the distribution of CC,TT + CT genotype of PLA2G2 A rs11677 C / T SNP was66. 2%( 172 /260) and 33. 8%( 88 /260),respectively. There was no significant difference in the positive rate of PLA2G2 A protein expression between cases with CC genotype( 56. 4%,97 /172) and cases with CT + TT genotype( 55. 7%,49 /88)( P〉0. 05). The genotypes rate of PLA2G2 A rs11677 C / T did not vary by age,histological differentiation,TNM stage,lymphatic metastasis and depth of invasion of ESCC( P〉0. 05). The positive rate of PLA2G2 A protein expression did not vary by age,histological differentiation,TNM stage and lymphatic metastasis( P〉0. 05). There was no significant difference in the positive rate of PLA2G2 A protein expression among ESCC patient groups with different rs11677 C / T genotypes( χ2= 0. 149, P =0. 928). Conclusion The correlation between PLA2G2 A rs11677 C / T polymorphism and the PLA2G2 A protein expression is not significant,while PLA2G2 A protein expression is closely related to the depth of invasion of ESCC.
