摘要
目的检测浙江温州及宁波地区献血人群中与输血相关性急性肺损伤(TRALI)密切相关的人类中性粒细胞抗原-3(HNA-3)的分布频率,以评估该地区献血人群中TRALI风险。方法共筛选来自浙江温州及宁波地区的献血者血液样本2 316例,采用通用基因组DNA提取试剂盒提取受试者外周血白细胞总DNA,并用Taq Man SNP基因分型探针法进行HNA-3基因分型实验,对基因分型为HNA-3a和HNA-3b的样本分别测序验证,用SPSS 16.0软件统计分析HNA-3等位基因频率。结果 2 316例献血者中共检出HNA-3a/3a有1 172例(50.6%,1 172/2 316),HNA-3a/3b有965例(41.7%,965/2 316),HNA-3b/3b有179例(7.7%,179/2 316),其中HNA-3a基因频率为0.714,HNA-3b基因频率为0.286。结论浙江省献血人群中具有较高的HNA-3b/3b纯合子携带比率,由此产生的TRALI潜在风险需引起临床和采供血机构的密切关注。
Objective The aim of this study was to investigate the frequency distribution of human neutrophil antigens-3(HNA-3) in blood donors of Wenzhou and Ningbo population,Zhejiang province,so as to evaluate the risk of TRALI. Methods Screened samples from 2 316 unrelated healthy blood donors from Wenzhou and Ningbo. The white blood cell total DNA of the donors were extracted by universal genomic DNA extraction kit. The Taq Man SNP Genotyping Assay was applied to HNA-3a and HNA-3b genotyping,then validated by gene sequencing and then SPSS 16 software was used for statistical analysis of HNA-3 allele frequency. Results 117 2 cases of HNA-3a /3a were detected in the 231 6 cases of blood donors(50. 6%,1 172 /2 316),965 cases of HNA-3a /3b( 41. 7%,965 /2316),179 cases of HNA-3b /3b( 7. 7%,179 /2316),where HNA-3a gene frequency was 0. 714,HNA-3b gene frequency was 0. 286. Conclusion In Zhejiang province there is high blood donation crowd HNA-3b /3b homozygote carrier rate,the TRALI potential risks resulting need paid close attention to clinical and blood collection agencies.
