The expression β-catenin and GSK-3β in different molecular typing of breast cancer and its significance

摘要 目的探讨β-catenin、GSK-3β在不同分子分型乳腺癌中的表达情况、机制及意义。方法收集214例乳腺癌,根据ER、PR、HER-2、Ki-67的表达情况进行分子分型,即Luminal A型、Luminal B型、HER-2过表达型和三阴型,并进行β-catenin、GSK-3β免疫组织化学染色,阐述β-catenin、GSK-3β在不同分子分型乳腺癌中的表达情况及意义。结果 214例乳腺癌中Luminal A型32例(14.95%),Luminal B型104例(48.60%),HER-2过表达型27例(12.62%),三阴型51例(23.83%),β-catenin在四种亚型中的异常表达情况分别为:Luminal A型(7/32,21.88%),Luminal B型(24/104,23.08%),HER-2过表达型(13/27,48.15%),三阴型(29/51,56.86%),组间比较差异具有统计学意义;GSK-3β在四种亚型中的表达情况分别为:Luminal A型(22/32,68.75%),Luminal B型(22/104,59.62%),HER-2过表达型(10/27,37.04%),三阴型(12/51,23.53%),组间比较差异均具有统计学意义;β-catenin与GSK-3β相关性分析显示,在Luminal型、Luminal B型、三阴型中两者呈负相关,在HER-2过表达型中两者无相关性。结论β-catenin、GSK-3β在不同分子分型乳腺癌中表达存在差异性,并且两者的表达存在相关性,机制与Wnt/β-catenin信号通路的激活及调控密切相关。 Objective We aimed to investigate the expression of β-catenin and GSK-3βin different molecular typing of breast cancer cases and its mechanism and clinical significance.Methods We collected 214 cases of breast cancer cases, they were divided into four subtypes according to the expression of ER, PR, HER-2 and Ki-67, namely Luminal A type, Luminal B type, HER-2 over expression type and triple-negative type.Immunohistochemical staining methods was employed to detect the expression ofβ-catenin and GSK-3β, further elaborated the expression ofβ-catenin and GSK-3βin different molecular typing of breast cancer and its significance.Results Among the 214 cases of breast cancer there were 32 cases of LuminalA type (14.95%), 104 cases of LuminalB type (48.60%), 27 cases of HER-2 over-expression (12.62%), 51 cases of triple-negative type (23.83%).The expression ofβ-catenin in the four subtypes were as LuminalA type (7/32, 21. 88%), LuminalB type (24/104, 23.08%), HER -2 over expression type (13/27, 48.15%), triple -negative type (29/51, 56.86%), the differences among the four group were statistically significant (P< 0.05). GSK3βexpression in the four subtypes were as LuminalA type (22/32, 68.75%), LuminalB type (22/104, 59.62%), HER -2 over expression (10/27, 37.04%), triple -negative type (12/51,23.53%), the differences among the four groups were statistically significant (P< 0.05).β-catenin and GSK-3βcorrelation analysis showed a negative correlation between the two groups.Conclusions The expression of β-catenin and GSK-3βexist in different molecular subtypes of breast cancer exist difference and relative correlation between their expressions may closely related to the mechanism of Wnt /β-catenin signaling pathway activation and regulation.
