Girdin蛋白在大肠癌组织中的表达及其与侵袭转移的关系 被引量:3

Expression of Girdin protein and its correlation with tumor's invasion and metastasis in colorectal carcinoma

摘要 目的 检测大肠癌组织中Girdin蛋白的表达,探讨与大肠癌临床病理特征及侵袭转移的关系.方法 应用免疫组织化学Envision二步法,对84例大肠癌及40例腺瘤、30例癌旁黏膜组织进行Girdin蛋白检测,统计分析其与临床病理参数之间的关系.结果 84例大肠癌组织中Girdin蛋白阳性表达率为42.9%,癌旁黏膜组织、低级别上皮内瘤变(LGIN)腺瘤及高级别上皮内瘤变(HGIN)腺瘤Girdin蛋白阳性表达率分别为3.3%、11.1%、18.2%,差异有统计学意义(x2=15.752,P<0.01;x2=6.391,P<0.05x2 =4.518,P<0.05).大肠癌组织中Girdin蛋白的表达与肿瘤浸润深度、淋巴结转移及临床分期明显相关(P<0.01),与性别、年龄、肿瘤大小、肿瘤部位、组织学分级无明显相关(P>0.05).大肠癌Girdin蛋白阳性表达组的5年生存率为30.6%,阴性组5年生存率为83.3%,两组间差异有统计学意义(P<0.01).结论 Girdin蛋白在大肠癌组织中呈高表达,与大肠癌的侵袭转移有关. Objective To investigate the expression of Girdin protein in colorectal carcinoma,and explore its relationship with clinical characteristics and tumor' s invasion and metastasis.Methods Eighty-four cases of colorectal carcinoma specimens,40 cases of colorectal adenoma and 30 cases of tumor-adjacent mucosal tissues were collected,and the expression levels of Girdin protein were detected by immumihistochemistry Envision two-step methods.The relationship between Girdin expression and clinicopathological characteristics was analyzed.Results The positive expression rate of Girdin protein in colorectal carcinoma was 42.9%,which was significantly higher than that in tumor-adjacent mucosa,adenoma with low grade inraepithelial neoplasia (LGIN) and adenoma with high grade inraepithelial neoplasia (HGIN) respectively (x2 =15.752,P 〈 0.01 ;x2 =6.391,P 〈 0.05 ;x2 =4.518,P 〈 0.05).Overexpression of Girdin protein was correlated with invasion depth,lymph node metastasis and Dukes clinical stage (P 〈 0.01),but not with gender,age,pathologic grade,tumor's size and localization (P 〉 0.05).Follow-up study results showed that the five-year overall survival rate in Girdin negative group (83.3%) was higher than that in Twist positive group (21.7%) significantly (P 〈 0.01).Conclusion Girdin protein expression is up-regulated in colorectal carcinoma and correlated with tumor's invasion and metastasis.
