乳腺癌中淋巴管生成与SOX2和血管内皮生长因子-C的关系 被引量:5

摘要 目的探讨乳腺癌中淋巴管生成与SOX2和血管内皮生长因子-C(VEGF-C)的关系,以及SOX2、VEGF-C、淋巴管生成与乳腺癌临床病理参数的关系。方法收集青岛市市立医院病理科2013年1月~2014年4月乳腺癌组织标本90例,采用免疫组化SP法检测各乳腺癌组织中SOX2、VEGF-C及Podoplanin的表达。比较分析不同临床分期及病理组织学分级的乳腺癌组织中SOX2、VEGF-C及淋巴管密度值(LVD)的表达情况。结果 90例乳腺癌组织中,淋巴结转移者SOX2、VEGF-C和LVD的阳性率分别为84.62%、76.92%和61.54%,高于无淋巴结转移者(44.00%、48.00%、32.00%),差异均有统计学意义(χ2=15.23、7.05、6.33,P=0.00、0.01、0.01)。组织学分级Ⅱ~Ⅲ级者SOX2、VEGF-C的阳性率分别为82.35%、76.47%,高于组织学分级Ⅰ级者(45.45%、45.45%),差异均有统计学意义(χ2=11.57、7.46,P=0.00、0.01)。SOX2、VEGF-C阳性者LVD阳性率分别为60.60%和61.29%,高于SOX2、VEGF-C阴性者(33.33%、35.71%),差异均有统计学意义(χ2=5.26、5.07,P=0.02、0.02)。SOX2阳性者VEGF-C阳性率为75.76%,高于SOX2阴性者(50.00%),差异有统计学意义(χ2=5.45,P=0.02)。结论 SOX2、VEGF-C、LVD阳性表达提示肿瘤的高侵袭性,且三者间均存在一定的相关性。SOX2在肿瘤中可能通过促进VEGF-C分泌诱导淋巴管生成。 Objective To investigate the correlations between lymphangiogenesis and SOX2,VEGF-C in breast carcinoma,and its associations with the clinical parameters of patients.Methods Immunohistochemistry SP method was applied to detect the expression of SOX2,VEGF-C and podoplanin in 90 cases of breast carcinoma from the Department of Pathology in Qingdao Municipal Hospital from January 2013 to April 2014.The expression of SOX,VEGF-C and LVD in tissues of breast carcinoma with different clinical stage and histologic grading were compared and analyzed.Results In 90 tissues of breast carcinoma,the positive rates of SOX2,VEGF-C and LVD in tissues of patients with lymph node metastasis were 84.62%,76.92 and 61.54% respectively,which were much higher than those of non-lymph-node-metastasis(44.00%,48.00%,32.00%),the differences were statistically significant(χ2=15.23,7.05,6.33;P=0.00,0.01,0.01).The positive rates of SOX2 and VEGF-C in tissues of patients with histological grade of Ⅱ-Ⅲwere 82.35% and 76.47% respectively,which were much higher than 45.45% and 45.45% in patients with histological grade Ⅰ,the differences were statistically significant(χ2=11.57,7.46;P=0.00,0.01).The positive rates of LVD were60.60% and 61.29% in patients with positive expression of SOX2 and VEGF-C respectively,which were much higher than patients with negative expression of SOX2 and VEGF-C(33.33%,35.71%),the differences were statistically significant(χ2=5.26,5.07;P=0.02,0.02).The positive rates of VEGF-C was 75.76% in patients with positive expression of SOX2,which was much higher than those patients with negative expression of SOX2(50.00%),the difference was statistically significant(χ2=5.45,P〈0.05).Conclusion The positive expression of SOX2,VEGF-C or LVD indicates highly malignant tumor and their expression levels are correlated.SOX2 may induce lymphangiogenesis by promoting the secretion of VEGF-C.
