摘要 目的探讨肝X受体(LXR)对糖尿病肾病大鼠的影响及机制。方法选取60只Wistar大鼠,采用抽签法随机分为对照组、模型组及干预组,每组各20只。模型组及干预组大鼠通过链脲佐菌素(60 mg/kg)腹腔注射建立糖尿病肾病模型,干预组大鼠给予LXR激动剂T0901317(30 mg·kg-1·d-1)。2周后,称重,留取24 h尿,之后腹腔注射乙醚,在腹上动脉部位进行取血,随后处死,留存血液样本,测量血糖、肾功能、血脂水平。结果 12周后,3组大鼠体质量、尿蛋白、血糖、肌酐、尿素氮、内生肌酐清除率、三酰甘油、总胆固醇、高密度脂蛋白胆固醇水平比较,差异均有统计学意义(P<0.05);其中模型组和干预组尿蛋白、血糖、肌酐、尿素氮、三酰甘油、总胆固醇水平高于对照组,体质量、内生肌酐清除率、高密度脂蛋白胆固醇水平低于对照组(P<0.05);干预组尿蛋白、血糖、肌酐、尿素氮、三酰甘油、总胆固醇水平低于模型组,体质量、内生肌酐清除率、高密度脂蛋白胆固醇水平高于模型组(P<0.05)。结论 LXR表达能够明显改善Wistar大鼠糖尿病肾病生化指标,具有一定的指导意义。 Objective To investigate the effects of liver X receptor( LXR) expression on rats with diabetic nephropathy( DN) and its mechanism. Methods 60 Wistar rats were randomly divided into control group,model group and intervention group by sortition randomization method with 20 rats in each group. Rats in model group and intervention group were injected intraperitoneally with streptozocin( STZ)( 60 mg / kg) injection to establish DN model. Rats in intervention group were treated with LXR agonists T0901317( 30 mg·kg- 1·d- 1). After 12 weeks,the rats were weighed up,24- hour urine of rats was collected,then rats were injected intraperitoneally with ether,blood samples were taken from epigastric vessels,then rats were put to death,the blood samples were used to measure levels of blood sugar,renal function,blood lipid. Results After 12 weeks,there were significant differences in body mass, levels of urine protein, blood sugar, creatinine, urea nitrogen,triglyceride,total cholesterol and high density lipoprotein cholesterol, and endogenous creatinine clearance rate among three groups of rats( P〈0. 05). Levels of urine protein,blood sugar,creatinine,urea nitrogen,triglyceride and total cholesterol in model group and intervention group were significantly higher than those in control group( P〈0. 05), while body mass,endogenous creatinine clearance rate,and level of high density lipoprotein cholesterol in model group and intervention group were significantly lower than those in control group( P〈0. 05). Levels of urine protein, blood sugar, creatinine, urea nitrogen,triglyceride and total cholesterol in intervention group were significantly lower than those in model group, while body mass,endogenous creatinine clearance rate,and level of high density lipoprotein cholesterol in intervention group were significantly higher than those in model group( P〈0. 05). Conclusion LXR expression can obviously improve biochemical indexes of Wistar rats with DN,which may have certain guiding significance.
