温阳活血解毒方对ApoE^(-/-)小鼠动脉粥样硬化斑块及斑块内血管新生的影响 被引量:14

摘要 目的探讨温阳活血解毒方对ApoE-/-小鼠动脉粥样硬化(AS)易损斑块及斑块内血管新生的作用。方法以高脂饲料喂养雄性ApoE-/-小鼠13周,复制AS小鼠模型。将模型小鼠随机分成模型组,辛伐他汀组,温阳活血解毒方低、中、高剂量组,分别给予蒸馏水、辛伐他汀和温阳活血解毒方干预13周。测定小鼠动脉斑块内成分(细胞外脂质成分、泡沫细胞、平滑肌细胞、胶原纤维),计算易损指数,并测定斑块内新生血管的密度。结果与模型组比较,辛伐他汀组及温阳活血解毒方中、高剂量组易损指数、斑块内新生血管密度均有明显降低(P<0.05)。温阳活血解毒方中、高剂量组对斑块易损指数、新生血管密度的作用优于低剂量组(P<0.05),而高剂量组对新生血管密度的作用优于中剂量组(P<0.05)。结论温阳活血解毒方可通过减少AS斑块内泡沫细胞、增加斑块内平滑肌和胶原成分,降低斑块内新生血管的表达以稳定AS斑块,并呈一定的剂量依赖性。 Objective To investigate the effects of Wenyang Huoxue Jiedu(WHJ) formula on the atherosclerotic vulnerable plaque and intraplaque angiogenesis of ApoE^-/-mice.Methods A total of 100 8-week male Apo E-/-mice were fed with high-fat diet for 13 weeks to induce atherosclerosis(AS).And then the modeled mice were randomly divided into model group,simvastatin group,low-dose WHJ group,middle-dose WHJ group,high-dose WHJ group.Distilled water,simvastatin and WHJ formula were given to the corresponding groups following the experimental design for another successive 13 weeks.The components of arterial plaque of each group,including extracellular lipid composition(ELC),foam cells(FC),smooth muscle cells(SMC),collagenous fibers(CF),were counted.The atherosclerotic plaque vulnerability index and new vessel density were also calculated.Results Compared with the model group,the vulnerability index and intraplaque microvessel density of the simvastatin group and middle- and high-dose WHJ groups were much decreased(P〈0.05),the decrease of middle- and high-dose WHJ groups was superior to that in simvastatin group.High-dose WHJ group had better effect on microvessel density than middle-dose WHJ group(P〈0.05).Conclusion WHJ formula shows an effect on stabilizing the AS plaque by decreasing intraplaque foam cells and microvessel density and increasing intraplaque SMC and CF,and the effect is dose-dependent.
