根皮苷抑制体外LO2肝细胞脂肪变性作用的实验研究 被引量:4

摘要 目的研究根皮苷对体外LO2肝细胞脂肪变性的抑制作用。方法体外培养LO2细胞,实验分正常对照组、模型组、辛伐他汀组和不同剂量的根皮苷组。药物作用24,48,72h后分别检测油红O染色后脂滴吸光度,细胞内甘油三酯(TG)含量和细胞外培养基葡萄糖水平。结果与模型组比较,光镜下各给药组的细胞脂滴均明显减少。在24,48,72h时模型组吸光度均显著高于正常对照组(P<0.01),各给药组吸光度均显著低于模型组(P<0.05,P<0.01);在24,48,72h时模型组TG水平均显著高于正常对照组(P<0.05,P<0.01),辛伐他汀组及根皮苷高、中剂量组TG水平均显著低于模型组(P<0.05,P<0.01);在24h时模型组细胞培养基上清液葡萄糖含量均高于正常对照组(P<0.05),各给药组葡萄糖含量均显著低于模型组(P<0.05,P<0.01)。结论根皮苷对油酸诱导的体外LO2肝细胞脂肪变性和糖代谢异常有显著的抑制作用。 Objective To observe the effect of phloridzin on inhibiting fatty degeneration of the human liver LO2 cells.Methods The in-vitro cultured LO2 hepatocytes were divided into several groups:phloridzin treatment groups with different concentrations of phloridzin(200,400,800 μ mol·L-1,respectively),simvastatin-treating group,model group and normal control group.After treatment with the corresponding agent for 72 hours,the changes of lipid droplets in the LO2 hepatocytes were detected by oil red O staining,and triglyceride(TG) content in hepatocytes and glucose level in the supernatant were measured by commercial kits.Results Compared to the normal control group,lipid droplets,oil red O value,and TG at culture hour 24,48,72,and glucose level at culture hour 24 were obviously increased in the model group(P〈0.05,P〈0.01).Phloridzin and simvastatin could decrease the intercellular accumulation of lipid droplets.Oil red O value in simvastatin group and high-,and low-dose phloridzin groups was lower,and glucose level in the supernatant of the four medication groups at culture hour 24 was also lower than those in the model group(P〈0.05,P〈0.01).Conclusion Phlorizin could suppress the abnormal changes in adipose degeneration and glucose metabolision induced by oleic acid in the LO2 hepatocytes.
